Mechanism Of Gelsemine Action,and It’s Effects On IPEC-J2 Cell Proliferation

Gelsemium elegans(G.elegans),also known as “Duan-Chang” grass,is a traditional Chinese medicine.G.elegans can cause ill-effects,including vomiting,diarrhea,severe abdominal pain and even death,if consumed accidently.Pigs and goats eat G.elegans,which will lead them to gain weight and shiny hair,G.elegans has rowth-promoting effects.Indole alkaloid is a component of G.elegans,such as koumine,gelsemine,gelsenicine,and they are relevant to growth-promoting ability.Objective: Gelsemine is effects on promote cell proliferation,providing evidence of G.elegans indole alkaloids growth-promoting effects.And in the experiment,intestinal porcine epithelial cells(IPEC-J2)was used as the research cell,to explore the mechanism of gelsemine promote animal cell proliferation.Methods: The experiment method such as: CCK-8,q RT-PCR,flow cytometry,Western Blot,5-Ethynyl-2’-deoxyuridine(Ed U)cell proliferation test.To detect the effects of gelsemine done on IPEC-J2 cells about cell viability,Gene m RNA expression,protein expression,cell proliferation ability.Results: IPEC-J2 cell viability is enhanced by gelsemine,which concentrations range in10 μM ～ 40 μM.10 μM and 20 μM gelsemine are the best effects.And gelsemine could promote IPEC-J2 cell proliferation in 10 μM and 20 μM concentrations(P < 0.05).Under the effect of gelsemine,the Epidermal growth factor receptor(EGFR)gene m RNA expression has been clearly increased in IPEC-J2 cells(P < 0.01).the protein of EGFR expression has been increased(P < 0.01),level of p-EGFR(Y1173)has been increased(P < 0.01).Meanwhile,gelsemine could decrease the Liver X receptors alpha gene promoter(NR1H3)m RNA expression(P < 0.05).Expression of extracellular regulated protein kinases 1/2(ERK1/2)has changed,the level of p-ERK(Thr202/Tyr204)and p-ERK(Tyr222/Tyr205)has increased(P <0.05).And enhance cell cycle transform speed,to prove cell proliferation rate.Conclusion: Gelsemine could promote the EGFR,EKR1/2 m RNA expression,and protein of EGFR expression.Gelsemine could enhance EGFR protein and ERK1/2 protein activity.Activating ERK1/2 MAPK pathway,promote cell proliferation.
