| Tabanusisan important blood-sucking insect,which is important for animal health and disease transmission as a vector.The study of mitochondrial genetic variation characteristics of tabanus accumulates basic data for exploring phylogenetic relationship of species and functions of different mitochondrial genes.In this study,samples from five different species were collected from a sheep farm in Xinyang,Henan province,and the genetic characteristics of their mitochondrial genomes were analyzed by high-throughput sequencing technology and bioinformatics methods.The results showed that:1.In this study,the mitochondrial genome lengths of the Tabanus wereobserved to be between 15847 bp and 16016 bp,encoding 13 protein-coding genes,22 t RNA genes,2 r RNA genes and 1 control region gene.The use of codons preference,evolution patterns and other aspects of different species were highly similar.These genes had the same sequence in their own mitochondrial genome.2.The content of AT in the mitochondrial genome ranges from 77% to 78%,which showed obvious AT bias.3.The genetic variation analysis of nad2,nad5,J-PCGs combination,N-PCGs combination,r RNA combination gene and amino acid combination sequence of PCGs werecarried out by bayesian method and maximum likelihood method,and it was found that these Tabanus all showed close relationship in the phylogenetic analysis of different gene combinations,and they were members of the same cluster of tabanidae.In addition,both Tabanus Yamasaki Ouchi and Tabanus mandarinus Schiner belonged to the Group amaenus and the results of this phylogenetic analysis werwhighly consisted with the subfamily and population classification.4.The results of the evolutionary tree obtained from different genes and gene combinations were different,and the reason may be that the evolution rate of different genes in different species was different,and the difference was also influenced by the effect of natural selection.The result indicated that the mitochondrial genome could be used in the biological classification and identification of tabanus,but it needed to be comprehensively investigated in the evolutionary analysis,rather than the genetic evolution of a single gene. |
