There are a lot of special sheep breeds in China,of which it has excellent characteristics such as,high reproduction,flavor,and stress resistance etc.However,these breeds in China have an obvious shortage that is low growth rate and products,and lead that it is difficult for meeting the demand of consumers.Some breeds have more meat quality was imported from all over the world to improve the meat quality of local Chinese sheep breeds,as a result the aim of improving meat quality gradually comes true.Dorper,a famous mutton sheep breeds for high products and meat quality,was used to improve the Chinese sheep meat quality in general.Dorper sheep always make changes to adapt the different natural environment in China and it may lead to the offspring possessing a bit of distinction in genes or protein expression,even physiological function,and that is the aim of exploring in this study.Male and female Dorper sheeps from Henan and Jilin province as research objectives to find out the differences in blood physical and chemical indicators and blood transcriptome sequencing the results were as follows:1.Blood physicochemical indicators between male and female Dorper sheeps in JilinThe physiological indexes of Dorper rams in Jilin area,such as percentage of monocytes(MON%),hemoglobin(HGB),mean hemoglobin(MCH),mean hemoglobin concentration(MCHC),ratio of large platelets(P-LCR),and biochemical indexes,such as total cholesterol(CHOL)and creatinine(CREA),were significantly higher than those of ewes(P<0.05).The biochemical indexes of Dorper rams in Jilin area,such as albumin(ALB),alanine transaminase(ALT),triacylglycerol(TG),The hormone indexes Neuropeptide Y(NPY),ghrelin,leptin,melanocortin,growth hormone(GH),insulin-like growth factor 2(IGF2),uncoupling protein 2(UPC2),uncoupling protein 3(UPC3)were significantly higher than those of ewes(P<0.01);The physiological indexes of Jilin Duper ewes,such as red blood cells(WBC),percentage of lymphocytes(LYM%),biochemical indexes,such as calcium(Ca)and low-density lipoprotein(LDL-C),were significantly higher than those of rams(P<0.05).The physiological indexes of Jilin Dorper ewes,such as lymphocytes(LYM),red blood cells(RBC),platelets(PLT),pro Calcitonin(PCT),mean platelet volume(MPV),biochemical indexes,total protein(TP),urea nitrogen(UREA)Glutamyltransferase(GGT),phosphorus(P)and high-density lipoprotein(HDL-C)were significantly higher than those of rams(P<0.01),and there was no significant difference in other indicators.2.Blood physicochemical indicators between male and female Dorper sheeps in HenanThe physiological index of Calcitonin(PCT)and biochemical index of urea nitrogen(UREA)of Dorper rams in Henan were significantly higher than those of ewes(P<0.01);The physiological index high-density lipoprotein(HDL-C)and hormone index Melanocortin of Henan Dorper ewes were significantly higher than those of rams(P<0.01),and the other indexes had no significant difference.3.Blood physicochemical indicators of Dorper rams in Henan and JilinThe physiological indicators of Henan Dorper rams,such as hematocrit(HCT),were significantly higher than those of Jilin Dorper rams(P<0.05).The physiological indicators of Henan Dorper rams,such as red blood cell(RBC),mean platelet volume(MPV),large platelet ratio(P-LCR),and biochemical indicators such as urea nitrogen(UREA),were significantly higher than those of Jilin Dorper rams(P<0.01);The percentage of monocytes(MON%),mean hemoglobin concentration(MCHC),platelet hematocrit(PDW),mean hemoglobin concentration(MCH)and biochemical index high-density lipoprotein(HDL-C)of Jilin Dorper rams were significantly higher than those of Henan Dorper rams(P<0.05).The physiological index mean hemoglobin concentration(MCH),biochemical index albumin(ALB)The total cholesterol(CHOL)was significantly higher than that of Dorper rams in Henan region(P<0.01),and there was no significant difference in other indicators.4.Blood physicochemical indicators of Dorper ewes in Henan and JilinThe physiological indexes,mean hemoglobin(MCH),mean hemoglobin concentration(MCHC),hormone indexes,Neuropeptide Y(NPY),ghrelin,leptin(LEP),melanocortin,growth hormone(GH),insulin-like growth factor 2(IGF2),uncoupling protein 2(UPC2),uncoupling protein 3(UPC3)of Henan Dorper ewes were significantly higher than those of Jilin Dorper ewes(P<0.01);The physiological indicators of red blood cell(WBC)and biochemical indicators of glutamyltransferase(GGT)in Jilin region Dorper ewes were significantly higher than those in Henan region Dorper ewes(P<0.05).The physiological indicators of platelet(PLT),biochemical indicators of urea nitrogen(UREA),and phosphorus(P)in Jilin region Dorper ewes were significantly higher than those in Henan region Dubo ewes(P<0.01),There was no significant difference in other indicators between Dorper rams and Dorper ewes in Jilin.5.Blood transcriptome of Dorper sheeps in Henan and JilinBy comparing the blood transcriptome sequencing results of Dorper sheeps in Henan and Jilin,225.36 Gb valid data were obtained,and 3530 differentially expressed genes(2170 upregulated and 1360 down-regulated)were screened.The differential genes were significantly enriched in 51 pathways(P<0.05),including 36 biological processes,9 cellular components,6molecular functions,and 1012 related genes.KEGG enrichment resulted in a significant enrichment of 2731 differently expressed genes(p.adjust<0.05)among 67 signaling pathways.Among them,the NOD receptor signaling pathway and JAK-STAT signaling pathway may be pathways that affect the immune and signal transduction of Dubo sheep.The expression levels of the main functional genes NOD2,NLRP1,MAVS,TRAF3,AKT,PI3 K,JAK1,and JAK2 in the pathway were significantly different between Dorper sheeps in the two regions,It is speculated that it may be the main gene affecting immunity and signal transduction.In summary,Dorper sheeps have strong adaptability in both Henan and Jilin regions.Although there are differences in blood physicochemical indicators,most of them are within the normal range,and some indicators exceeding the normal range may be caused by stress reactions;The results of blood transcriptome sequencing showed that the adaptability to the outside world was different due to different growth environments in different regions,which caused changes in the expression of immune related genes in the body to adapt to the environment.The research results of this article have laid a theoretical foundation for further exploring genes related to stress resistance in meat sheep and exploring immune regulation mechanisms.The differently expressed genes obtained can provide genetic materials for the breeding and improvement of Dorper sheeps in the later stage,and also provide a large amount of basic data for exploring genes that affect the adaptability of the body,providing important significance for promoting reasonable hybridization and industrial upgrading of meat sheep in China. |
