Escherichia coli and Staphylococcus aureus are common pathogens in veterinary clinic.At present,antibiotics are mainly used in veterinary clinical prevention and treatment,but the frequent use of antibiotics will increase the drug resistance of bacteria,which increases certain difficulties for clinical treatment.Chinese herbal medicines have a long history,good therapeutic effect,little toxic and side effects,wide sources,no residual and other advantages.In this study,30 Chinese herbal medicines were first screened out with good antibacterial effect on E.coli and S.aureus,and then compound screening was conducted according to the screening results.Drug safety test and mouse treatment test were conducted on the obtained Chinese herbal compound.To provide basic data for the development and application of TCM compound in clinic.1.In this study,30 kinds of Chinese herbs were selected,and the effective components were extracted from 30 kinds of Chinese herbs by decocting in water and extracting in alcohol respectively.The antibacterial effects of the extracts on E.coli and S.aureus were measured,and the single Chinese herbs with good antibacterial effects were comprehensively screened out according to the antibacterial circle diameter,MIC and MBC.The results showed that the Chinese herbs with good bacteriostasis against E.coli and S.aureus were: sea buckthorn,scutellaria scutellaria,pomegranate peel,phelloderma phelloderma,wormwood leaf,tangerine peel and licorice.2.According to the screening test results of single flavor of Chinese herbal medicine,7 flavors of Chinese herbal medicine had significant antibacterial effect on E.coli and S.aureus.The 7 flavors of Chinese herbal medicine were combined by orthogonal test,and the compound preparation with the concentration of 1 g/m L was prepared.The results showed that the best herbal compound Ⅶ was pomegranate peel: scutellaria scutellaria: Sea buckthorn: Mugwort leaf(4:1:16: 4),the inhibitory zone diameter,MIC and MBC of Compound Ⅶ were 23.67 mm,7.81 mg/m L and 15.62 mg/m L for E.coli,and22.00 mm,1.95 mg/m L and 3.91 mg/m L for S.aureus,respectively.3.Kunming mice were divided into 1 group control group and 3 groups experimental group.The experimental groups were intragaadministrated with high(2 g/m L),medium(1 g/m L)and low(500 mg/m L)concentration of Compound Ⅶ extract solution,and were intragaadministrated continuously for 5 days.The pathological sections of stomach,small intestine,liver and kidney of mice in the high concentration group were the same as those of the control group,and the survival rate of mice in all groups was 100%.Therefore,the maximum safe concentration of Compound Ⅶ was 2 g/m L,indicating that compound Ⅶwas a safe preparation without poison.In the experimental treatment of mice infected with E.coli and S.aureus,the effect of compound Ⅶ and cefquinme sulfate injection was the same,and the cure rate reached80%.The above results showed that seabuckthorn,pomegranate peel,scutellaria scutellaria,Phelloderma phelloderma,licorice,wormwood leaves and tangerine peel had obvious antibacterial effects on E.coli and S.aureus.Ⅶ : Pomegranate peel: Scutellaria scutellaria: Sea buckthorn: Mugwort leaf(4:1:16:4).Compound Ⅶ is safe and non-toxic,and the therapeutic effect of compound Ⅶ is the same as that of the antibiotic cefquinme sulfate injection in mice.It can be used as an antibiotic substitute in the treatment of E.coli and S.aureus infectious diseases in veterinary clinic,and provides reference for the follow-up clinical trials.
