Isolation And Identification Of Pathogenic Bacteria From Diarrhea Lambs In Large-scale Sheep Farms And The Efficacy Of Drug Control

Abstract/Summary:
With the continuous development of the meat and sheep industry,it’s scale and breeding number are also increasing,and healthy lamb breeding is an important step to ensure the economic benefits of sheep farms.However,lambs have low immunity and are extremely sensitive to changes in the external environment,which is easy to cause various diseases.Among them,lamb diarrhea as the most common disease,the cause is complex,improper treatment will lead to a large number of lamb death,directly affect the economic benefits of sheep farms.In this study,an intensive sheep farm in Dingyuan County where lamb diarrhea is frequent was selected to determine the microflora of lamb feces through bacterial isolation and identification,drug susceptibility test and clinical treatment effect analysis,and at the same time to determine the microflora of lamb feces,in order to propose the best treatment plan for lamb diarrhea in this farm.1.Bacterial isolation and identification: Feces of 20 lambs with diarrhea were collected.Isolation and culture,biochemical reaction and nucleic acid identified pathogens as E.coli and C.Verichner.Verichneri was the pathogen of lamb diarrhea,while E.coli was a nonpathogenic bacterium.Drug susceptibility testing using the tablet method found that C.virneri was highly sensitive to ampicillin,ampicillin,chloramphenicol,tetracycline,and others.2.Clinical treatment effect: taking untreated diarrhea lambs as the control(D),the remaining 32 diarrheal lambs were treated with ampicillin sodium(Ta),whitehead oral liquid(Tb)and ampicillin sodium + white-headed lamb oral solution(Tab)for therapeutic observation trials,and the results showed that the average treatment days and average diarrhea index of lambs under the three drugs were 2.25~3.50 days and 4.50~11.25,respectively,with the Tab group having the best treatment effect and the Tb group having the worst treatment effect.Further analysis of blood immunological indexes showed that compared with group D,the contents of bacterial lipopolysaccharides(LPS)and LPS connexin(LBP)in the blood of lambs(P < 0.05)were significantly reduced in the three drug treatment groups,while the contents of blood interleukin-10(IL-10),IL-1β,melatonin(MT)and catalase(CAT)were significantly higher than those in the untreated diarrhea group(P < 0.05),and the LBP and IL-1β contents were the lowest in the Tab group.3.Effect of clinical treatment on fecal flora: Tb and Tab groups significantly increased microbial community richness in lambs(P < 0.05),while the Ta group were not significantly different from untreated diarrhea(P > 0.05).At the phylum level,the relative abundance of Fusobacterium in feces of lambs was significantly increased compared with Group D,at the genus level,the relative abundance of Fusobacteria,Helicobacter,Parasutterella,Parasutterella,Enterococcus,and Helicobacter was significantly higher than that in Group D(P < 0.05),while the relative abundance of Corynebacterium and unclassified_Bacilli was significantly lower than that in Group D(P < 0.05).In addition,the richness of the microbial community in the Tab group showed the highest relative abundance of Fusobacterium,Fusobacterium,unclassified_Bacilli,and snail species.In summary,the pathogen causing lamb diarrhea in the sheep farm was Clostridium weisseri,and the clinical treatment effect was the best treatment effect of ampicillin sodium +albicanium oral liquid,which provided a reference for further in-depth research.
