Abstract/Summary:
In this study,weaning lambs were taken as the research object to explore the effects of adding medical stone carrier compound artemisia seed polysaccharides and organic acid preparations to the diets on the glucose and lipid metabolism and intestinal health of weaned lambs,in order to develop new compound additives.This research is divided into the following four parts:Experiment 1: Using the in vitro gas production method,the appropriate addition ratio of artemisia seed polysaccharides(ASP)and organic acid in the diet of weaned lambs was determined through the test of dietary fitness and the influence of in vitro fermentation parameters and degradation rate.The results of the study found that the addition of ASP compound additives could promote rumen fermentation,improve the rumen environment,increase the degradation rate of dietary dry matter and organic matter,and improve the efficiency of microbial biosynthesis to a certain extent.Among them,medical stone 5g/d +ASP 5g/d,medical stone 5g/d + ASP 5g/d + glycerol monolaurate 3g/d,medical stone 5g/d+ ASP 5g/d + fumaric acid 5g/d + disodium fumarate 5g/d,medical stone 5g/d + ASP 5g/d+ coated sodium butyrate 5g/d is the most suitable,as the additive doses of T1,T2,T3 and T4 group respectively in the following animal experiments.Experiment 2: The effect of adding ASP compound additives in the diet on the nutritional metabolism and blood indexes of weaned lambs.30 Alxa cashmere goat weaned male kids with similar body weight(21.55±2.43kg)were selected and divided into 5groups on average,CK group(basic diets),T1,T2,T3 and T4 group.The pre-trial period was 7 days,and the positive trial period was 50 days.The body weight of the experimental sheep was weighed every 10 days.At the end of the animal experimentation,3 kids of each group slaughtered,a total of 15 kids.The results showed that each experimental group had a trend of increasing kid weight gain and slaughter rate.The addition of T3 and T4 groups increased the apparent digestibility of DM,NDF and CP(P<0.05).),the T4 group increased the apparent digestibility of organic matter(OM)(P<0.05),and the T1 group increased the apparent digestibility of CP(P<0.05).The contents of GC in the T1 group and T3 group were significantly higher than those in the CK group(P<0.05),the contents of TC and LDL-C in the T1 group and T2 group were significantly decreased(P<0.05),and the contents of T-AOC and SOD in the T1 group were significantly increased(P<0.05).0.05),Ig M content in T1,T2,T3,and T4 groups was significantly higher than that in CK group(P<0.05),Ig A content in T1 group was significantly higher than that in CK group(P<0.05),and FGF19 was increased in T2 and T4 groups.The content of FGF21 increased in T4 group.Experiment 3: The effect of adding ASP compound additives to the diet on the expression of intestinal health-related genes in weaned lambs.The results showed that adding ASP compound additives increased the m RNA expression of intestinal barrier-related genes claudin-1,claudin-2,Occludin,ZO-1,and the m RNA expression of GLUT2 gene in ileum tissue was significantly higher than that of CK group,but lower in cecal tissue(P<0.05).The T2 group significantly increased the m RNA expression of PPAR-α gene in the duodenum,and T4 significantly decreased the m RNA expression of AMPK gene in the cecum(P<0.05),and promoted the expression of IL6 in the ileum,TNF-a in the colon,and IL6 in the duodenum(P<0.05).All experimental groups inhibited the expression of NF-κB in the liver,T3 and T4 groups increased the expression of ALB in the liver,and T4 decreased the expression of HP.Experiment 4: The effect of adding medical stone carrier compound formulation to the diet on the shape of the intestinal villi of weaned lambs.The test results showed that the jejunum of T1,T2,and T3 groups and the villus height was higher than that in the CK group(P<0.05),and the ratio was also higher than that in the CK group(P<0.05).The overall conclusions of the experiment are as follows:(1)The addition of ASP and organic acid preparations can promote rumen fermentation,improve rumen internal environment,and increase feed dry matter degradation rate.(2)The addition of ASP compound additives and monolaurin compound to the diet of weaned lambs can reduce the content of TC and LDL-C in serum,which is beneficial to the growth and development of weaned lambs.And increased the content of T-AOC and SOD in serum,have antioxidant function,and improve the immune function of animals.(3)ASP compound additives and sodium butyrate can reduce intestinal permeability to a certain extent and maintain intestinal epithelial barrier function.It can promote intestinal transport and absorption and intestinal glucose and lipid metabolism in lambs,and has a certain inhibitory effect on intestinal and liver inflammation.(4)Supplementation of ASP and organic acid preparations in the diet could improve the villus height of jejunum and ileum,the ratio of jejunum-intestinal villus height / crypt depth,and ileal crypt depth.
