The total flavonoids were extracted by ultrasonic,and then eluted by gradient of ethanol with different concentrations through polyamide chromatography colμmn.The eluted components of FLAVONOIDS were 35%,55%,75% and 95%.The total flavonoids have biological functions such as antioxidant,anti-inflammatory,anti-tμmor,anti-bacterial,enhancing body immunity and improving animal growth performance.However,there are few studies on different concentrations of ethanol eluting components of flavonoids of S.amurensis.In view of this,this thesis aims to explore the inhibitory effect of different concentrations of ethanol elution fractions on salmonella typhimuriμm,Bacillus cereus,Staphylococcus aureus,Escherichia coli and other common bacteria by in vitro antibacterial test.Mice feeding experiment was conducted to explore the effects of different concentrations of ethanol elution components on immune system,antioxidant system and intestinal morphological and structural indexes in mice,so as to provide theoretical research for further development and utilization of different concentrations of ethanol elution components and the application of related health products of R.mongolicμm.Ninety Kunming mice(6-8 weeks old)were random Ly divided into 9 groups with 10 mice in each group.The control group was intragastric to sterilize tap water,and the experimental groups were intragastric at 60 mg/kg BW·d and 120 mg/kg BW·d respectively,eluting 35%,55%,75% and 95% of AMinoflavone.Results of this test:In experiment 1,the antibacterial effects of different flavones on Salmonella typhimurium,Bacillus cereus,Staphylococcus aureus and Escherichia coli in vitro were investigated.The test results show that: 0.625 mg/ ml,1.25 mg/ ml,2.5 mg/ ml and 5 mg/ml had certain inhibitory effects on the above four kinds of bacteria,and the order of inhibitory effect was as follows: Bacillus cerus > Salmonella typhimurium > Escherichia coli > Staphylococcus aureus.In addition,the 55% ethanol eluting fraction had more significant antibacterial effect,which was superior to 35%,75% and 95% of the four kinds of bacteria.The antibacterial zone diameter was ≥8 mm,especially for Bacillus cereus.The maximum antibacterial zone diameter was 15 mm when the concentration was 5 mg/ML.In experiment 2,the effects of different flavones on immune indexes and cytokines secreted by immune cells in mice were studied.The results showed as follows: the 35% eluting fraction of 120 mg/kg BW·d could significantly increase the immune canal index of mice(P<0.01),and the 95% eluting fraction of 120 mg/kg BW·d could significantly increase the thymus index of mice(P<0.01).Different elution fractions of 60mg/kg BW·d and 120 mg/kg BW·d could improve the levels of immunoglobulin Ig(A,M,G)in mice,among which 35% elution fraction of 120 mg/kg BW·d and 95% elution fraction of 120 mg/kg BW·d had the most significant effect on immunoglobulin Ig(A,G)in mice.The 75% elution fraction of 120 mg/kg BW·d had the best effect on immunoglobulin Ig(M)in mice.The 95% elution fraction significantly increased the levels of IFN(α,γ)and TUMOR necrosis factor-α(TNF-α)in spleen tissues of mice(P<0.01),and the 35% elution fraction had the best regulation effect on IFN(β).In summary,95% eluting components had the best effect on various immune indexes and cytokines secreted by immune cells in mice,followed by 35% eluting components.In experiment 3,the effects of different flavones on antioxidant function of mice were investigated.The test results show that: 35% elution fractions of 60 mg/kg BW·d and 120mg/kg BW·d could significantly reduce the content of MDA in liver tissues of mice(P<0.01).35% and 55% elution fractions of 60 mg/kg BW·d,120 mg/kg BW·d could significantly increase the levels of CAT and T-SOD in liver tissues of mice(P<0.01).The concentrations of 60 mg/kg BW·d and 120 mg/kg BW·d of different flavones could increase the level of GSH-Px in liver tissues of mice.In conclusion,the results of 35%elution group of 120 mg/kg BW·d were better than 55%,75% and 95% groups.In experiment 4,the effects of different flavones on intestinal morphology and structure of mice were studied.The results showed that 60 mg/kg BW·d and 120 mg/kg BW·d of different flavones could increase the villus height of duodenum,jejunum and ileum of mice.The crypt depth of duodenum,jejunum and ileum was decreased.The(V/C)ratio of duodenum,jejunum and ileum was enhanced.Among them,35% elution fraction of 120 mg/kg BW·d and 95% elution fraction of 120 mg/kg BW·d had the best effect. |
