Sugar beet,a biennial herbaceous crop of Chenopodiaceae,is one of the main sugar crops in China,which is mainly grown in Northwest and North China such as Inner Mongolia and Xinjiang.However,the above-mentioned areas are relatively scarce in water resources,with large seasonal differences in precipitation,and some areas still restrict agricultural water use.Drought has become the main factor restricting the development of sugar beet production.Glycosyltransferases(GTs)are enzymes that catalyze the transfer of donor glycosyl groups to specific receptors to generate glycosidic bonds,and play an important role in response to a variety of biotic and abiotic stresses.The previous studies of our research group has shown that the sugar beet GTs gene Bv UGT90A1 had obvious response to drought stress.In order to clarify the role of Bv UGT90A1 in sugar beet drought resistance,this gene was cloned from drought resistant sugar beet variety HI0466,analyzed by bioinformatics,subcellular localization and drought stress expression.Plant overexpression vector was constructed and transformed into Arabidopsis to identify its function.The results showed that:1、The full length of CDS was 1960 bp,including a 1464 bp open reading frame,encoding 487 amino acids,and its fat coefficient was 86.86.The encoded protein was predicted to be a hydrophobic unstable protein.2、Subcellular localization showed that it was mainly located in cytoplasm and nucleus;Phylogenetic tree analysis showed that its encoded amino acids were closely related to those encoded by the flax mustard glycosyltransferase gene Cs UGT90A1 and the Gynostemma.glycosyltransferase gene Gp UGT90A1.3、QPCR analysis showed that the expression of this gene in Sugar beet leaves was significantly up-regulated under drought stress,indicating that it could respond positively to drought stress.The identification of drought resistance in transgenic Arabidopsis showed that the Bv UGT90A1 gene could increase the activity of SOD,POD and CAT and the content of total flavonoids,improve the antioxidant capacity,reduce the level of MDA and maintain the membrane stability,and then improve the drought resistance of plant.
