The rhinoceros beetle,known by its scientific name,the double-fork rhino golden turtle,which is distributed in Zhejiang and other provinces in China,and belongs to the genus rhinoceros beetle of the order Hymenoptera and.With a long-time recordation in the literature that male rhinoceros beetle featuring efficacy in relieving fear,pain,stasis and toxicity,there was few studies that have been conducted in the utilization as medicine.Given the huge damage of forest sparkled by swarms of it,and stick to rechange it to treasure,a study of aqueous extract of male rhinoceros beetle was conducted for its analgesic effect on mice,the safety evaluation and nutrient analysis.The contents of amino acids,fats and fatty acids in male rhinoceros beetles were determined given the national standard method for the aim of analysing the nutritional composition of male rhinoceros beetle.While the results revealed that the crude fat content of it was 13.77%,containing 16amino acids and 11 fatty acids,with 8 essential amino acids among those,accounting for 38.42%of the total amino acids;5 saturated fatty acids,accounting for 42.31%of the total fatty acids;and unsaturated fatty acids,accounting for 57.61%of total fatty acids.The male rhinoceros beetle were tested for harmful heavy metals and microorganisms given the national standard method for the aim of determining the food safety.While the results revealed that the levels of all harmful heavy metals in male rhinoceros beetle did not out of limit and contained beneficial trace elements as well.On the other hand,the results for Escherichia coli,Staphylococcus aureus,moulds and yeast in the tests for microbial pathogens were all negative and the total number of bacterial colonies,1.9 x 10~4 CFU/g,also pass muster the national food safety requirements.Aiming to study the analgesic effect of the aqueous extract of rhinoceros beetle on mice,the male rhinoceros beetle powder was studied by manner of aqueous extract,and ICR mice were divided into five groups featuring three groups of aqueous extract of male rhinoceros beetle containing low(0.13g/m L),medium(0.26 g/m L),high(0.52 g/m L)concentrations,positive drug(aspirin)control group and blank control group.Meanwhile,the analgesic effects in mice were investigated by the acetic acid torsion method,the hot plate tesk and the tail-flicking tesk in mice.Compared with the blank control group,the results revealed that the aqueous extracts of rhinoceros beetle in low,medium and high concentrations all served significantly vell in reducing the frequency of torsions(P<0.05),sinking the pain quotient(P<0.05)and prolonging the time of pain threshold(P<0.05)in the hot plate and tail-flicking tesks,which given play to analgesic effect on mice to some extent.In conclusion,containing rich amino acids and fatty acids in vivo,the male rhinoceros beetle is free from heavy metal contamination and microbial pathogens with an analgesic effect on mice of the aqueous extract.Therefore,the male rhinoceros beetle is a kind of resource insect featuring high nutritional value,safety and no toxic side effects. |
