| Objective: 1,Based on surveys that Solomon’s seal germplasm resources in anhui province,combing Solomon’s seal plant trait in different areas in anhui province and the suitability,for Solomon’s seal planting in different areas in anhui province and provide evidence for reasonable development and utilization.2,By comparing phenotypic traits and DNA molecular marker technology,the combination of analysis of different areas in anhui province Solomon’s seal genetic germplasm resources between groups3,Respectively by monosaccharide composition in the polysaccharide and aminoacids,nucleosides and chemical composition for the comprehensive evaluation index,different areas in anhui province based on the flower more diligenceMethods:(1)The characteristics of Polygonatum cyrtonema Hua in different are as were observed by field collection and data inquiry,and the information of 132 samples were collected,43 ecological factors were integrated,and the maximu m entropy model was used to analyze the growth suitability of Polygonatum cyrtonema Hua in different areas of Anhui Province.(2)Using 96 Polygonatum cyrtonema Hua germplasm as material,DNA was extr acted by CTAB method,and then amplified by PCR according to the reported SSR primers.According to the number of bands,10 pairs of primers with high polymorphism were selected and the data matrix was obtained.The polymorphism information content(PIC)of different primerloci was calculated by allele composition,and the genetic diversity and genetic structure of different populations were inferred by Gen Al Ex6.503 and Power Maker.(3)Polygonatum cyrtonema Hua polysaccharide was extracted by water extraction and alcohol precipitation,prote in was removed by Sevage method,hydrochloric acid was used to hydrolyze the polysaccharide,PMP(1-phenyl-3-methyl-5-pyrazolone)was used to derive the hydrolysate,and UHPLC-QTRAP-MS/MS was used to detect monosaccharide derivatives.The quality of Polygonatum cyrtonema Hua from different producing areas was comprehensively evaluated by TOPSISI method and PCA analysis.(4)the water extract of Polygonum polygonatum was extracted by ultrasonic,and 38 kinds of standard samples with different chemical components were analyzed by UHPLC-QTRAP-MS/MS instrument and the content of the samples was determined.Two multivariate statistical methods,TOPSIS and PCA analysis,were used to evaluate the quality of Polygonatum cyrtonema Hua in different producing areas.Results:(1)There were a large number of cultivated areas in Dabie Mountains and Southern Anhui Mountains,and there were significant differences in plant morphology between Dabie Mountains and Southern Anhui mountainous areas.the main environmental factors affecting the ecological suitability of Polygonatum cyrtonema Hua in Anhui Province are February precipitation,March precipitation,December precipitation and soon.In addition,the highly suitable areas of Polygonatum cyrtonema Hua in Anhui Province are Qingyang County,Shitai County,Ji nzhai County,Qimen County and other places.(2)A total of 70 alleles were det ected in Polygonatum cyrtonema Hua germplasm samples,and the optimal grouping number K was predicted to be 3,that is,Polygonatum cyrtonema Hua germplasm resources were mainly divided into three populations,namely,Jinzhai Polygonatum population,Qingyang Polygonatum population and Jingde Polygonatum population.Through the analysis of Fst and φ St values among populations,Polygonatum cyrtonema Hua and the other two populations have produced moderate genetic differentiation.(3)A method for rapid detection of different monosaccharides in Polygonatum cyrtonema Hua polysaccharides was established.Six monosac charides(mannose,arabinose,glucose,ribose,rhamnose,galactose)were detected.Among the monosaccharide components of Polygonatum cyrtonema Hua polysac charides,the content of mannose was the highest in polygonatum polysaccharides,with an average proportion of 67.71%,followed by glucose,galactose,arabinose,rhamnose and the lowest ribose content was only 0.31%.(4)To establish a method for rapid detection of 38 chemical components in Polygonatum cyrtonema Hua.The results showed that tyrosine,glutamic acid,leucine and serine were abundant in amino acids of Polygonatum cyrtonema Hua.The contents of guanosine and uridine in nucleosides were higher,and apigenin and diosgenin were more in other components.The analysis shows that the contents of amino acids and other chemical components in Polygonatum cyrtonema Hua are related to the geographical environment,and the contents of most components vary greatly in different regions.In the analysis results,the comprehensive evaluation of Polygonatu mcyrtonema Hua was higher in Qingyang area(S5)and Jinzhai area(S8),while cluster analysis showed that the comprehensive evaluation of Wuwei County(S2)and Tongcheng City(S3)in Anhui Province was lower than that in Tongcheng City(S3).Conclusion: The morphological characteristics of rhizome,leaf and peduncle of Polygonatum cyrtonema Hua have been different in different areas of Anhui,and the highly suitable areas of Polygonatum cyrtonema Hua in Anhui are Qingyang County,Shitai County,Jinzhai County,Qimen County and soon.The germplasm resources of Polygonatum cyrtonema Hua are divided into three clusters.The first cluster is mainly Polygonatum cyrtonema Hua in Jinzhai and Huoshan areas,the second cluster mainly contains Polygonatum cyrtonema Hua in Qingyang and Shitai,and the third cluster mainly includes Polygonatum cyrtonema Hua in Jingde,Huangshan and Qimen areas.the results are consistent with the results based on morphological classification.There are differences in morphology and genes,and the experimental results show that Qingyang population has high genetic diversity.Through the study on the contents of monosaccharides,amino acids and nucleosides in Polygonatum cyrtonema Hua polysaccharides,the results of comprehensive evaluation are consistent with the results of different regions in Anhui Province according to the suitability in the first chapter,Suitability region is higher than low suitability,high
