| Artemia nauplii and adults are important biological bait for the cultivation of seawater seedlings,taking advantage of the non-selective filter-feeding properties of Artemia.Through the way of nutritional enrichment,the intestines of Artemia are wrapped and carried with specific nutrient substances,which can effectively meet the needs of aquatic animals for essential fatty acids,amino acids,vitamins,etc.This study intends to use β-carotene as an enhancer by observing the effect of β-carotene enrichment on the composition and content of carotenoids in Artemia,to explore the change rule of carotenoids in Artemia.To evaluate the feasibility of Artemia as a carotenoid bio-carrier and to mine genes related to β-carotene metabolic transformation through transcriptomics.In order to explore the effect of β-carotene enrichment on the composition and content of carotenoids in Artemia,this paper took Artemia in Bohai Sea Gulf as the research object and selected four different concentrations of β-carotene(0,5,15,25mg/ L),to compare the changes of carotenoid composition and content in Artemia nauplius and adults after 24 h of enrichment and 24 h of starvation after enrichment,respectively.The results showed that during the enrichment process,the enriched β-carotene content of Artemia nauplius reached the highest at 12 h,the β-carotene content of Artemia adults reached the highest at 6h.The β-carotene content in Artemia nauplius increased significantly with the increase of the enriched dose.(P<0.05),showing an excellent enrichment effect;according to the results of the enrichment experiment,Artemia nauplius and adults were selected to be starved after 12 h and 6h of enrichment,respectively,and the stability of β-carotene in Artemia was evaluated.During the 24 h starvation process,the β-carotene content in Artemia decreased significantly with the time(P<0.05),and reached the minimum at 24 h,but the deposition of β-carotene in the high-dose enhanced group was still significantly higher(P<0.05)than in the low-dose enhanced group.In the study of the carotenoid composition of Artemia,after β-carotene enrichment,it was found that only three carotenoids in Artemia nauplius and adults,β-carotene,canthaxanthin,and echinenone.β-carotene enrichment had a significant effect on carotenoid composition in Artemia,and the changes of canthaxanthin in Artemia nauplius and adults also showed a significant correlation with the gradient ofβ-carotene fortification.During the enrichment process,the contents of echinenone and canthaxanthin in Artemia nauplius and adults increased significantly(P<0.05)and showed a trend of first increasing and then decreasing with the increase of enrichment time;the content of echinenone in both Artemia nauplius and adults decreased significantly after starvation(P<0.05),the content of canthaxanthin in Artemia nauplius showed a trend of first increasing and then decreasing(P<0.05).There was no significant difference compared to 6 h starvation with after enrichment(P>0.05),and it decreased significantly after 6h.Therefore,it is speculated that Artemia nauplius and adults can convert β-carotene into echinenone and canthaxanthin.The 0 mg/L β-carotene as the control group(CG)and 25 mg/L β-carotene as the treatment group(TG),the transcriptome sequencing of Artemia nauplius enriched for12 h was performed,a total of 42.44 Gb Clean Data was obtained,and the Clean Data of each sample reached more than 6.63 Gb,the percentage of Q30 bases is more than93.86%,the number of unigenes assembled is: 58202,the number of Transcriptions is:93648,using six major databases(NR,Swiss-Prot,Pfam,COG,GO and KEGG database)for comparison.Differential expression analysis was performed on the control and treatment group,and the results showed that 1279 genes were up-regulated and 1426 were down-regulated.The differentially expressed genes were subjected to GO and KEGG enrichment analysis.Finally,the critical enzyme genes and transcription factors involved in the metabolism and transport of β-carotene in Artemia were excavated. |
