Tianjin bohai bay A.sinica is a kind of high-quality biological bait,tolerance,hatching features and the content of nutrients can match the United States Saline Lake species.In recent years,due to the environmental pollution and other kinds of A.sinica introduction and applications,some kinds of A.sinica might have spread to Tianjin bohai bay making biological invasion to local A.sinica resource,so it is necessary to research and protect Tianjin bohai bay A.sinica.The aim of this thesis is to research and protect the resource of Tianjin bohai bay A.sinica.The major research contents include that: identifying the species of Tianjin bohai bay A.sinica;the effect of external environment factors on hatching characteristics and The influence of exogenous neurotrophic factor on its nutrients.The main resμLts are:1.Using DNA barcode technology to amplify COI sequence of Tianjin bohai bay A.sinica and analyze gene sequence by blast,the similarity is as high as 99% with COI sequence of Artemia parthenogenetica in Gen Bank.so the Tianjin Bohai Bay A.sinica is named Artemia parthenogentica BHW.2.Among three kinds of environmental factors,it is salinity that effect diapause cysts most.Experimental resμLts show that bohai bay A.sinica hatching salinity is 10‰-25‰,the best salinity is 25‰.Temperature has obvious effect on diapause cysts hatching.The hatching rate is highest when temperature up to 28±1 ℃.What this thesis differ from other studies is that under 36 ℃,some egg can still hatch,but under 44 ℃,few egg can hatch but can not live long time.Light have no significant influence on hatching rate.The best condition is 90μmol/(m2 · s).Three kinds of environmental factors have certain interaction,the optimal comprehensive environmental conditions of Tianjin bohai bay A.sinica is that light intensity is 72μmol/(m2 · s),salinity is 25‰,pH is 8.3.Eight kinds of exogenous neurotrophic factor can significantly improve protein content of A.sinicaNauplius after reinforcement.P.subcordiformis has the best effect.After reinforcement,crude fat has been improved significantly.N.closterium f.minutissima group is higher than any other group.Eight kinds of exogenous nutrients has no significant effect on the composition of amino acids in Artemia parthenogentica BHW.After strengthened the content of saturated fatty acid,monounsaturated fatty acid,polyunsaturated fatty acids and high unsaturated fatty acid is significantly improved than control group,C.Pyramidomona group is higher than other groups.Chaetoceros spand D.salina can significantly improve EPA of A.sinica,I.galbana can improve the content of DHA.High unsaturated fatty acids such as EPA and DHA are easy lack of,but in the aquacμLture seedlings its growth rate,disease resistance,survival rate,growth and reproduction has irreplaceable effect.So combined with aquacμLture production practice that the effect of I.galbanais the best,followed by Chaetoceros.sp and D.salina. |
