| Objective: With the intensive breeding conditions of livestock and poultry and the further development of modern breeding industries,the diarrhea rate of broken benchmarks guided by immune stress has continued to increase.The results of the preliminary test prove that Taurine(Tau)plays an important intervention role in the process of alexic diarrhea induced by lipopolysaccharide(LPS).This experiment plans to pass the 16 S rDNA sequencing and metabolic group prove that LPS is interacted with the intestinal microbial group and its metabolites and taurine during inducing cecum damage.The protein(Tight junction,TJ)effectively improves LPS-induced cecum barrier damage.Methods: Eighteen weaned piglets(28±3 days old),duroc × Landrace ×Yorkshire,with a body weight of 6.5±0.5kg,were selected in a two-factor treatment design.The experiment was divided into three groups with six replicates in each group,including the C group(basal daily dose + normal saline injection),L group(basal diet +LPS intravenous injection),and LPS+ Taurine group(LT group,basal diet+LPS intravenous injection +0.3% taurine).Daily feed intake,daily gain,and feed/meat ratio were recorded.The damage of the cecum of piglets by LPS and taurine was observed by transmission electron microscope.The expressions of ZO-1,Occluding,and Claudin-1 were detected by Western Blot and q RT-PCR.The bacterial composition of cecal contents of weaned piglets was detected and analyzed by 16 S rDNA sequencing.Results: Results: after adding 0.3% taurine to the diet of piglets injected with LPS intravenously,the daily gain,daily feed intake and feed meat ratio increased significantly.It can be seen that taurine has a certain effect on increasing daily gain,daily feed intake,and reducing feed meat ratio;The results of transmission electron microscopy showed that LPS induced severe cecal injury in weaned piglets,and taurine could significantly improve the intestinal injury mediated by LPS;LPS significantly decreased the expression levels of protein and m RNA of intestinal tight junction ZO-1,Occluding and Claudin-1(P<0.01),but this situation was completely reversed when taurine was added to the basal diet,and the expression levels of protein and m RNA of Occludin,Claudin-1,and ZO-1 increased significantly(P<0.01).The results showed that taurine had a protective effect on LPS mediated barrier injury;Compared with Con group,the richness,and diversity of fecal flora in the LPS group were significantly higher(P<0.01),and the relative abundances of Spirochaetota,Bacteroidetes,Bacteroidia,Negativicutes,Bacteroidaceae and Prevotellaceae were significantly higher(P<0.05),and the relative abundances of Lactobacillus,Ruminococcaceae and Firmicutes were significantly lower(P<0.05).The metabolites in LPS group included N-Lactoylethanolamine phosphate、Melatonin、D-Pantethine、Cyclocalamin 、 6-Chloro-N-(1-Methylethyl)-1,3,5-triazine-2,4-diamine and2-(1-Methylpropyl)-4,6-dinitrophenol decreased significantly compared with CON group(P < 0.01).However,taurine added to the diet of piglets injected with LPS completely reversed this effect and effectively alleviated the cecal injury induced by LPS.Conclusion: In the cecum model of weaned piglets induced by LPS,taurine can improve the intestinal environment by regulating the composition of microorganisms and the content of metabolites in feces,effectively reducing the injury of tight junction protein by LPS,and significantly improve the intestinal injury of piglets. |
