Resveratrol(RSV),a plant polyphenol,affords antioxidant,antiinflammation and other various biological functions.The purpose of this study was to investigate the effect and mechanism of RSV on gut microflora,expression of inflammation related genes and tight junction protein in pigs.It was studied by feeding RSV to Duroc×Landrace×Yorkshire pigs and lipopolysaccharide(LPS)-induced IPEC-J2 cells inflammatory model.It provides a scientific basis for developing RSV as a feed additive that can improve the gut health of pigs.The main research results are as follows:1?The effect of RSV on the gut microflora of pigs.The twelve Duroc×Landrace×Yorkshire pigs with an average weight of about 65 kg were randomly divided into two groups,each with 6 pigs,which were the control group(feeding basic diet)and the resveratrol group(RSV,feeding basic diet+400 mg/kg resveratrol).The average body weight of pigs were about 110 kg for slaughter test.The rectal contents of each pig was sequenced for 16 S r DNA analysis and RSV antibacterial test was carried out in vitro.The results showed that:(1)the addition of RSV can change the gut microflora of pigs.There is a tendency to increase the relative abundance of gut microflora such as Bacteroidetes and Lactobacillus,and extremely significantly reduce the relative abundance of conditional pathogenic microflora Streptococcus(P<0.05).(2)The bacteriostasis study showed that RSV extremely significantly inhibits the proliferation of Streptococcus suis(P<0.01)and inhibits the formation of biofilm.2?Effect of RSV on the expression of inflammation-related genes and tight junction protein in the gut of pig.(1)RSV was feeded healthy Duroc×Landrace×Yorkshire pigs,RSV significantly reduced the levels of serum inflammatory factors in pigs(P<0.05).RSV may inhibit the activity of the NF-?B signaling pathway and the expression of downstream inflammatory genes MIP-2,VCAM1,COX-2 and MIP-1? of NF-?B signaling pathway,thereby reduced the expression of porcine gut inflammatory factors TNF?,IL-6,IL-8 and IL-1?.While increased the expression of tight junction protein ZO-1 and Claudin-3 in the gut of pig.The results showed that RSV can reduce the occurrence of gut inflammation,enhance the gut barrier function and improve gut health in pigs.(2)LPS induced inflammation in IPEC-J2 cells,adding RSV effectively could inhibit the activity of NF-?B signaling pathway,reduced the expression of inflammatory cytokine genes in cells.RSV significantly increased the expression of tight junction protein in cells(P<0.05).The results showed that RSV can alleviate inflammatory response of IPEC-J2 cells by induced LPS.As a whole,RSV improved the gut microflora richness and diversity of pigs,increased the relative abundance of beneficial bacteria and inhibited the growth of Streptococcus suis.RSV reduced the expression of inflammation related genes in gut tissues of pigs and increased the expression of tight junction protein,thus improve gut health of pigs.
