| The purpose of this study was to investigate the effects of probiotic lignocellulosic(Opti Cell)on the mucosal immune barrier of cecum in chickens of different breeds.In this study,108 healthy local Bian Chickens and ISA Brown Chickens of the same age and weight were selected to be raised in a cage.The design of 2 × 3 factors was adopted: each chicken was randomly divided into 3 treatments,each treatment was 6 replicates,and each repeat was 6 chickens.During the incubation period(1-8 weeks),the control group was fed a basal diet,treatment 1(OL)and treatment 2(OH)added 1% and 2% Opti Cell to the basal diet.During the incubation period(9-20 weeks),a self-prepared feed with isoprotein was prepared.Treatment 1(OL)and 2(OH)contained 2% and 4% Opti Cell,respectively.During the egg laying period(21-50 weeks),the control group was fed a basal diet,treatment 1(OL)and treatment 2(OH)were added with 1.5% and 3%.The pre-feeding period was 1 week and the formal period was 50 weeks.The results show that:(1)At 20 weeks,the index of thymus immune organs in Bian Chickens was significantly higher than that ISA Brown Chicken(P<0.01),and the expression levels of TNF-α and IL-6 m RNA in Bian Chickens were lower than that ISA Brown Chicken.The expression lever of IL-10 m RNA in Bian Chickens was significantly higher than that ISA Brown Chicken(P<0.05).(2)Add Opti Cell for ISA Brown Chicken had no significant effect on spleen,thymus and bursa of fabricius immune organ index at 8 weeks,but OL group significantly reduced the V/C,and promote the inflammatory cytokines IL-6 m RNA expression(P<0.01),significantly reduced the IL-1 m RNA expression(P<0.05),significantly increased the anti inflammatory cytokine IL-10 m RNA expression(P<0.05).Add Opti Cell had no significant effect on V and V/C at 20 weeks,but had a significant effect on C,mucus layer thickness,TNF-α,IL-1,IL-10,IL-6,MUC2 m RNA expression,and OL group significantly increased the mucus layer thickness and IL-10,MUC2 m RNA expression(P<0.01),significantly reducing the C and TNF-α m RNA expression(P<0.01),significantly reduced the cecum IL-6 and IL-1 m RNA expression(P<0.05).Add Opti Cell had significant effect on cecum V/C,cecal mucous layer thickness,TNF-α,IL-1,IL-10,IL-6,MUC2 m RNA expression,and OL group significantly increased the thickness of cecal mucous layer(P<0.05),significantly increased the V/C and the expression of anti-inflammatory cytokines IL-10 and MUC2 m RNA(P<0.01),and significantly reduced the expression of pro-inflammatory cytokines TNF-α,IL-1 and IL-6 m RNA(P<0.01).(3)At 20 weeks,the interaction between the Opti Cell and the breed had significant effects on the thickness of the mucus layer and the expression of the pro-inflammatory cytokines TNF-a and MUC2 m RNA(P<0.05).At 50 weeks,the interaction between probiotic lignocellulosic had an extremely significant effect on the thickness of the cecum mucous layer and the expression of IL-10 m RNA(P<0.01),and a significant effect on the expression of MUC2 m RNA(P<0.05).In summary,long-term addition of low-fiber group can improve the mucosal immune barrier of the cecum of ISA Brown Chicken and Bian Chicken. |
