Hazelnut is one of the four major dried fruits in the world.In this study,phenotypic traits and iPBS markers were used to analyze the genetic diversity of 36 Ping’ou hybrid hazelnuts collected.The genetic diversity of 35 phenotypic traits of 36 hybrid hazelnut varieties(lines)was analyzed,and the coefficient of variation and genetic diversity index of each trait were calculated and F test was carried out.SPSS 21.0 and SAS V8 were used for correlation,principal component and cluster analysis,and the Ping’ou hybrid hazelnuts were comprehensively evaluated.The genetic diversity and genetic relationship of 37 Ping’ou hybrid hazelnuts were analyzed by using 10 iPBS markers,and the genetic similarity calculation,UPGMA clustering analysis and PCA analysis were carried out by NTSYSpc 2.10 e software.The main research results are as follows:1.The genetic variation of 36 tested materials is very rich,the genetic diversity index of 35 phenotypic traits ranged from 0.237 to 2.098,with the highest genetic diversity index of fruit longitudinal diameter(2.098)and the lowest genetic diversity index of leaf back hair(0.237).2.The results of correlation analysis show that the correlation among all traits is complex,and most of them show extremely significant correlation and significant correlation,among which the extremely significant positive correlation coefficient between nut transverse diameter and nut longitudinal diameter is the largest,which is 0.959;The negative correlation coefficient between petiole thickness and nut lateral diameter is the largest,which is-0.265.Principal component analysis showed that the cumulative contribution rate of the first 11 principal components was 84.00%,and 85-202 had the best comprehensive characters.3.The tested materials were analyzed from 10 iPBS polymorphic primers.The PIC of 10 primers ranged from 0.38 to 0.50,with an average of 0.46.The genetic similarity coefficient of 35 Ping’ou hybrid hazelnuts amplified by iPBS-PCR ranged from 0.3243 to 0.9684,,and the threshold difference was 0.6441.Among them,the genetic similarity coefficient between materials 80-8 and 84-14 was the smallest,which was 0.3243,indicating that they had the farthest genetic relationship and the greatest variation.The genetic similarity coefficient between materials 85-69 and 84-12 is the highest,which is 0.9684,indicating that they have the closest genetic relationship and the smallest variation.4.Based on UPGMA clustering results,35 Ping’ou hybrid hazelnuts can be divided into 6 groups at the genetic similarity of 0.6952,and all 35 germplasms can be distinguished at the genetic similarity coefficient of 0.9450.Cophenetic correlation test r=0.7949,which shows that the clustering results are good.The results of UPGMA cluster analysis are consistent with principal component analysis(PCA).The above results indicated that the tested Ping’ou hybrid hazelnuts materials were rich in genetic diversity,and iPBS technology could be used to study the genetic diversity of Ping’ou hybrid hazelnuts.This study provides a reliable reference for the effective utilization of Ping’ou hybrid hazelnuts germplasm resources and the selection of parents in the breeding of superior species. |
