Exp.1 Effects of Feeding Broad bean on Flesh Characteristics in Zebrafish Based on MetabolomicsTo investigate the effects of dietary broad bean on flesh characteristics of zebrafish and the acting mechanism,a compound feed(control group)and soaked broad bean(broad bean group)were fed to zebrafish with initial body weight of(0.22±0.01)g for 60 days.The results showed that weight gain,survival,muscle fiber area and sarcomere length of broad bean group were significantly lower(P<0.01),whilethe muscle collagen content and muscle fiber density were significantly higher than those of the control group(P<0.01).The metabonomics showed that there were fifteen metabolites significantly changing in the muscle of broad bean group when compared to the control group,including eplerenone hydroxy acid,trimethylamine,Gly-His-Lys,formoterol,acetylcholine,γ-aminobutyric acid,acetylcarnitine,1,2-dipentadecanoyl-sn-glycero-3-phosphocholine,alanine betaine,erucic acid,stachytine,pantothenic acid,maltotriose,glycerophosphocholine and L-carnosine.Dietary broad bean mainly affected the fatty acyl metabolites and the metabolic pathways including unsaturated fatty acid biosynthesis and linoleic acid metabolism in muscle.Above all,broad bean had a negative effect on the growth of zebrafish,but it could improve the flesh characteristics.The acting mechanism may be related to the fatty acyl metabolites in muscle,and the fifteen metabolites can be used as potential markers.Exp.2 Effects of Feeding Broad bean on Muscle of Zebrafish Based on TranscriptomicsThis study investigated the effects of feeding soaked broad bean on muscle transcriptomics of zebrafish.Compared with the control fish fed with formula diet,the fish fed with soaked broad bean showed a significant change in genes expression in muscle.A total of 1166 differentially expressed genes(DEGs)were identified,including930 up-regulated DEGs and 1453 down-regulated DEGs.GO annotation showed that DEGs were mainly enriched in three GO terms as cellular process,metabolic process and biological regulation.KEGG annotation found that up-regulated DEGs mainly affected the signaling pathways of folding,sorting and degradation,and down-regulated DEGs mainly affected the signaling pathways of lipid metabolism,carbohydrate metabolism and amino acid metabolism.KEGG pathway enrichment analysis displayed that there were five pathways were significantly enriched in broad bean group,including oxidative phosphorylation,PPAR signaling pathway,cardiac muscle contraction,proteasome and glycolysis/glycosylation.Broad bean might promote the synthesis of collagen in muscle fibroblasts by inhibiting oxidative phosphorylation pathway,and improve fibrosis of muscle fibroblasts and collagen content by inhibiting the PPAR signaling pathway and reduce the length of sarcomere by inhibiting the path of myocardial contraction.
