Scabiosa tschiliensis was a perennial herbaceous flower,as well as an important wild medicinal plant.It has been wildly used in the garden due to its beautiful and colorful flower,as well as highly cold resistance,drought resistance and barren resistance.Scabiosa tschiliensis also has anti-inflammatory,antipyretic,antioxidant,sedative and immune-enhancing effects.However,with the shortage of natural resources in Scabiosa tschiliensisa,the market was in short supply.Therefore,the use of plant tissue culture and breeding technology can not only meet the market demand,but also be of some significance to protect its germplasm resources.This paper by Scabiosa tschiliensis planting seedlings leaf as the experimental material,Studied the best disinfection method of explants,the effect of plant growth regulators on the induction and differentiation,proliferation,cultivation,rooting and transplanting,and established the rapid propagation system of Scabiosa tschiliensisa.The test results were as follows:1.The best sterile treatment of explant was: first soak them in 75% alcohol for30 s,then soak them in 2% sodium hypochlorite for 9 mins.This disinfection method was the best,and the survival rate was 90.00%.2.The optimal medium for callus induction was MS+2.00 mg/L TDZ,the induction rate was 83.33%,and the multiplication ratio was 3.60.3.The optimal medium for inducing callus differentiation was MS+1.00 mg/L6-BA,the induction rate was 88.50%,and the proportion of adventitious buds larger than 3.00 cm was 36.00%.4.The optimal medium for inducing adventitious bud proliferation was MS+0.80mg/L 6-BA,the induction rate was 92%,and the proportion of the number of adventitious buds larger than 3.00 cm was 44.00%.5.The optimal medium for strong seedlings was MS+0.60 mg/L 6-BA,the induction rate was 96.00%,and the proportion of adventitious bud number larger than3.00 cm was 61.00%.6.The optimal rooting medium was 3/4 MS medium.7.The most suitable matrix components for transplanting regenerated seedlings were peat soil,perlite and sand,with a volume ratio of 4:3:2.The survival rate of transplanting was 80.00%,and the seedlings grew well. |
