Feather color is an important characteristic of poultry breed,which is directly or indirectly related to economic useage and production performance.In order to investigating deeply of the molecular markers related to the feather color traits,this paper takes the F1 generation of Sichuan white geese and Lande geese as the research materials,and divides the feather color phenotype into three types according to different parts.Three genes,kit,Rab27 A and MITF,were taken as candidate genes,and sequenced by Sanger Sequencing technology to scan the SNPs and correlated with feather color,which provided reference for feather color genetic research and breeding practice of geese.The main results are as follows:1.Four SNPs were found in KIT gene of geese,one of which was on exon 19(c.2526C>G),which lead to amino acid changes(ser839cys),the rest three SNPs were on exon 22(c.3311C>T,c.3507A>G,c.3544T>C).c.2526C>G was highly significantly correlated with the overall feather color phenotype of geese(P<0.01);3311C>T was highly significantly correlated with the white feather and spotted feather in the overall feather color of geese(P<0.01);c.3507A>G was highly significantly correlated with the gray feather and white feather in the overall feather color of geese,and was significantly correlated with the melanin diffusion defect in the wing feather area and the integrity of white stripe in the chest(P<0.05);3544T>C was highly significantly correlated with the overall feather color of geese(P<0.01),there was a significant correlation between the type and the relative character of melanin diffusion defect in wing feather area(P<0.05).According to haplotype analysis,7 haplotypes and 14 haplotypes combinations were obtained,among which the dominant haplotypes and dominant haplotypes were K2(CCAT)and K2K2,respectively.The correlation analysis showed that the haplotypes were highly significantly related to the overall feather color phenotype of geese(P<0.01),and significantly related to the integrity of the chest white stripe(P<0.05).2.Three SNPs(c.2653C>A,c.2795G>A,c.2903A>G)were found in exon7 of RAB27 A gene,all of which were located in 3’-UTR.Among them,the c.2653C>A and c.2903A>G loci were highly significantly correlated with the relative characteristics of white feather and spotted feather of the whole feather color of geese(P<0.01),and were highly significantly correlated with the relative characteristics of wide band and narrow band in the chest white band(P<0.01);the c.2795G>A loci were not significantly correlated with the feather color.Through haplotype analysis of the SNPs of RAB27 A gene,three haplotypes and three haplotype combinations were obtained,among which the dominant haplotype and dominant haplotype combinations were R1(CGA)and R1R1,respectively.The correlation analysis showed that the haplotypes combinations were significantly related to the white feather and spotted feather relative characters of the overall feather color of geese,and the wide band and narrow band relative characters of the chest white band(P<0.05).3.Three SNPs(c.1313C>T,c.1466C>T,c.1679A>G)were found on exon10 of MITF gene in geese,all of which had no amino acid change.Among them,the c.1313C>T locus was highly significantly related to the relative characteristics of gray white feather and full gray feather in wing feather area(P<0.01);the c.1313C>T locus was significantly related to the unilateral and bilateral gray white feather in wing feather area(P<0.05);the c.1466C>T locus was highly significantly related to the overall feather color phenotype,gray white feather and full gray feather in wing feather area(P<0.01);the c.1679A>G locus was significantly correlated with the gray feather and white feather relative traits of the overall feather color of geese(P<0.01).Through haplotype analysis of SNPs of MITF gene,7 haplotypes and 13 haplotypes combinations were obtained,among which the dominant haplotypes and dominant haplotypes combinations were M2(CCG)and M1M2,respectively.The correlation analysis showed that the haplotypes were highly significantly correlated with the feather area phenotype of goose wing(P<0.01).To sum up: KIT,RAB27 A and MITF genes may play an important role in the regulation of the overall feather color of geese.To be more specific,MITF gene plays a major role in the regulation of feather color traits in the wing feather region of geese,and RAB27 A gene plays a major role in the regulation of feather color traits of goose breast white stripe.In addition,KIT gene also plays an certain role in the regulation of feather color traits in wing feather region and chest white stripe.Based on the result,three genes may have gene interaction,so it is necessary to further explore the formation mechanism,so as to provide theoretical basis for feather color inheritance and breeding of geese. |
