To investigate the effects of different exercise modes and exercise time on the male fertility in healthy young mice,32 male C57BL/6J mice were randomly divided into 4groups: control group(CON,8);Exhaustive exercise(EE,8);Acute aerobic exercise(AAE,8);Chronic aerobic exercise(CAE,8).After the exercise training,the serum,testis,epididymis samples were collected.The serum testosterone were detected by ELISA;q PCR and HE staining were used to detect the spermatogenesis maker genes expression in testicular tissue and seminiferous lumen area and the shape of the epididymis canal and the abnormal sperm rate.The serum testosterone level of EE group was lower than that in group CON.Then,the elderly mice were selected for long-term aerobic exercise,and the elderly inactive mice were used as the control group.The serum testosterone levels of each group were detected by ELISA.HE staining and fluorescence quantitative PCR were used to analyze the differences in the lumen area and spermatogenic marker genes of testosterone convoluted spermatozoa,as well as sperm morphology.At the same time,boars were also selected as the research model to compare the total number of sperm of motile and immotile boars,and sperm motility and sperm deformity rate were detected by microscopic examination.Finally,metabolomics was used to compare the changes of serum metabolites before and after longterm aerobic exercise and inactivity in male rats.The results showed that:1)The relative expression of in STAR m RNA of group CAE were significantly lower than that in group CON,and the lumen area of seminiferous tubule and the abnormal sperm rate were higher than that in group CON(P<0.05).The phenotypes of mice in the AAE group were the same as the CON group.The serum testosterone level of CAE group was higher than that in group CON.The relative expression of in STAR m RNA of group CAE were significantly higher than that in group CON,and the lumen area of seminiferous tubule and the abnormal sperm rate were lower than that in group CON(P<0.05).2)compared with the control group,long-term aerobic exercise can significantly increase the total number of sperm in the aged mice(P <0.05),reduce the rate of sperm malformation(P < 0.05),significantly reduce the testicular convoluted fine tubule cavity area(P < 0.05),improve the shape of the epididymis tube,and thus improve the reproductive ability of the aged mice.3)Compared with the control group,long-term aerobic exercise can significantly increase the total number of spermatozoa,increase sperm motility and reduce the rate of sperm deformity(P < 0.05).4)Compared with the control group,long-term aerobic exercise can significantly activate the signaling pathways related to the metabolism of fatty acids and amino acids in mice,promote the metabolic pathway of tricarboxylic acid cycle,and increase the content of intermediate metabolites in the tricarboxylic acid cycle.In conclusion,CAE could effectively improve the male fertility in mice.AE could ineffectively improve the male fertility in mice.EE was harmful to the male fertility in mice.
