Grifola frondosa is scientifically named as Polyporus Bayanus,also known as Lotus Mushroom,Cloud Mushroom,Chestnut Mushroom,Maitake Mushroom,and Chestnut Mushroom.It is an edible and medicinal fungus.The meat is tender,crisp,delicious,Popular in Japan and North America.Grifola frondosa,as a medicinal fungus,can activate human immune function,thereby fighting cancer,stabilizing blood pressure,and can also be used as an anti-diabetic drug.The cultivation history of Grifola frondosa in our province is relatively short,the cultivation technology is not yet mature,and there are still many technical problems in production.At present,the degradation of strains in Grifola frondosa production has become one of the important issues restricting the development of Grifola frondosa industry.In this experiment,the purification and rejuvenation technology of Grifola frondosa and the fruiting mode were studied.The main research results are as follows:1.The method of tissue separation,cutting-edge separation and protoplast regeneration was used to purify and rejuvenate Grifola frondosa strains.The effects of three purification and rejuvenation techniques were compared.The results showed that:the method of tissue separation was used to purify and rejuvenate Grifola frondosa strains.The effect is the best,the mycelium grows the fastest,2.997mm/d,which is significantly faster than the control,and the mycelium is white and dense,with neat edges.2.Three kinds of bag placement methods are used in the fruiting period,including short bag vertical,short bag inverted and short bag vertical scratching.The results show that the fruit body of Grifola frondosa has agronomic characteristics,nutritional quality,and growth.Cycle,biological efficiency,and yield indicators have a significant impact.Among them,the suitable placement method for Grifola frondosa fungus sticks is to treat with scratching bacteria before covering the soil and then place it in a vertical position.The effect is the best,which shows that the fruit body has good agronomic characteristics and nutritional quality.It is better,the growth cycle is short,the yield is the highest,and its biological efficiency reaches 86.91%,which is 8.56%higher than the control.3.Through the study of different cover soil thickness of Grifola frondosa,the suitable cover soil thickness for Grifola frondosa cultivation is selected as 1cm,which is characterized by short growth cycle,highest yield,good agronomic characteristics of the fruit body,and both the content of crude polysaccharides and crude fat.It is the highest,its biological efficiency reaches 84.5%,which is 6.15%higher than the control.4.The fruiting effect of different soil heights during the cultivation of Grifola frondosa with half-covered soil was studied.The results show that the half-covered soil treatments are not as good as the control.The biological efficiency of the control is the highest,reaching 78.35%,which is higher than the half-covered soil height treatment.20%~40%.5.Using vermiculite and soil as soil covering materials,the effects of different soil covering substrates on the growth cycle of Grifola frondosa,biological efficiency,agronomic characteristics and nutritional quality of the fruit body were studied,and the soil covering substrate suitable for Grifola frondosa cultivation was selected as " 75%vermiculite+25%soil",that is,the ratio of vermiculite to soil is 3:1,the yield is the highest,and the biological efficiency reaches 104.73%,which is 26.38%higher than the control.6.The suitable period for spraying water-soluble fertilizers containing amino acids was studied.The results showed that the most suitable period for spraying was to treat the Grifola frondosa bag before fruiting,the yield was the highest,and its biological efficiency was 103.42%,which was higher than the control 25.07%.7.The effect of spraying ferrous sulfate on the growth and development of Grifola frondosa studied.By spraying different concentrations of ferrous sulfate at the budding stage and coral stage of Grifola frondosa,the suitable spraying stage for Grifola frondosa was selected.When the iron concentration is 0.35mmoL/L,the fruit body has good agronomic characteristics,round florets and high nutritional quality.The weight of a single mushroom is 1.51kg,and the yield is 10.07kg;the suitable iron concentration for spraying during the Grifola frondosa period was selected.When it is 0.4mmoL/L,the weight of a single mushroom is up to 1.47kg,and the yield is 9.7kg.The fruit body has good agronomic characteristics,round florets and high crude polysaccharide content.
