| Methyl jasmonate(MeJA)is a small molecular signaling substance with simple structure but very active in plants,which is involved in the induction of plant,fruit and vegetable resistance and other important reactions.Arginine,as the amino acid with the lowest C/N,is not only the nutrient of nitrogen storage,but also plays an important role in the stress resistance of plants and the improvement of postharvest quality of horticultural products.At present,the regulation of arginine synthesis and metabolismss by MeJA has been deeply studied in plant field,but few studies have been done in the field of edible mushrooms.In this study,Agaricus bisporus,the model material of edible mushrooms,was used to study the regulation of MeJA in the arginine biosynthesis and metabolisms during postharvest storage of Agaricus bisporus.The discovery in this study will provide a theory base for solving postharvest quality of Agaricus bisporus and exploring the role of arginine biosynthesis and metabolisms in the regulation of MeJA.The proteomics based on iTRAQ technology was used to identify 808 differential proteins related with the postharvest cap of Agaricus bisporus.Differential proteins related to arginine synthesis and metabolisms were selected to verify genes and proteins expression levels.These results indicated that the arginine synthesis and metabolismss were involved in the cap,and polyamines may be the main reason quality retention of Agaricus bisporus.Then,the genes and proteins expression levels related with arginine synthesis and metabolismss by the regulation of MeJA was detected.The results showed that MeJA regulated the expression of AbARG,AbOAT,AbASL,AbODC,AbSPDS,AbSPE-SDH and AbExpansin genes.The results of protein expression levels showed that MeJA regulated the expression of AbASL,AbARG,AbASS and AbSPDS protein.These results indicated that the role of arginine synthesis and metabolisms involves in MeJA-regulated postharvest quality retention of button mushroom.In different tissues(cap,gill and stipe),the genes and proteins expression levels related with arginine synthesis and metabolisms is as follows:At the genes and proteins expression levels,MeJA regulated the AbASL and AbSPDS proteins in three tissues.At the genes expression levels,MeJA regulated the AbARG、AbODC、AbOAT and AbDID2 genes in three tissues;MeJA regulated the AbExpansin gene in gill and stipe;MeJA regulated the AbSPE-SDH gene in cap and stipe;MeJA regulated the AbSAMDC gene in cap and gill;the AbASS and AbSAMS genes were regulated by MeJA in cap and stipe,respectively.At the proteins expression levels,MeJA regulated the AbASS and AbSAMS proteins in three tissues,while the AbARG proein in cap.These results indicated that the tissue-specific regulation of arginine synthesis and metabolisms by MeJA during postharvest storage of Agaricus bisporus.In conclusion,MeJA regulated the expression levels of genes and proteins related with arginine synthesis and metabolisms during postharvest storage of Agaricus bisporus.Besides,the regulation of AbASL,AbASS,AbARG,AbSPDS proteins and AbODC,AbOAT genes expression levels by MeJA were the most obvious. |
