In order to research the change law of anniversary of the testis and related gene expression in the process of sperm production,based on the conventional paraffin tissue section method,observed the third anniversary of chengdu and around area eel testis development situation,tested the eel bcas2 testis in different development period,dmc1,spo11 and rec8 expression,and uses the immune histochemical method to locate the position of eels dmc1 protein.The main research results are as follows:1.There were 454 male samples of 1627 eels collected from April 2015 to March 2018.The average length of male individuals was 38.53±5.46 cm(20.50~57.63cm),and the average weight was 50.70±24.07 g(17.70~143.30 g).The body length is mainly distributed in the range of 30.00~ 44.90 cm,and the body weight is mainly distributed in the range of 20.00~ 49.90 g.The total male rate of samples was 27.90%,and the peak appeared in March to April,July to September and November to December.The male rate in summer was significantly higher than that in winter(P <0.05),showing an obvious seasonal change rule.The sexual index fluctuated between0.18% and 1.31%,and the two peaks appeared in April and September of each year.Combined with histological observation,the peak reproduction time was determined in May of each year.It can be seen that the breeding period of rice eel in chengdu and surrounding areas is in April and September of each year,and the breeding peak is in May of each year.2.The expression of finches bcas2,dmc1,spo11 and rec8 in different developmental stages was detected by real-time quantitative PCR.The results showed that the expression trend of the four genes was basically the same,and the relative expression levels in the stage Ⅲ testis were significantly higher than those in stage Ⅱ and Ⅵ(P <0.05),while the expression levels were not significantly different in other stages of development(P >0.05).This suggests that bcas2,dmc1,spo11 and rec8 genes play an important role in the development of finches.3.Western blot results showed that Dmc1 positive signal could be detected in stage Ⅲ testis.Immunohistochemical results showed that the positive signal of Dmc1 was only located in primary spermatocytes.The results showed that Dmc1 protein may play an important role in the development of spermatocyte in rice eel. |
