Abstract/Summary:
Bombyx mori is a kind of insect with important economic value.Its colleterial gland is the place for secretion and storage of mucus.The substances secreted by the CGs have strong adhesion.The gluing substances play an important role in fixing and protecting the eggs,and provide the most suitable environmental conditions for larval hatching.In this study,analysis of comparative transcriptome was carried out using the pupae and moth of the P50 strain,and the sequencing results were analyzed by differential expression and enrichment.The results are as follows:1.Transcriptomic analysis of the pupae and moth of the P50 strain,we obtained a large number of differentially expressed genes.A total of 2136 differentially expressed genes were annotated for functional annotation,with 1213 up-regulated genes and 923 down-regulated genes.2.Based on GO annotation analysis of differentially expressed genes,we identified a large number of chitin binding and cuticle structural.Transporter activity was significantly enriched in molecular function,many transferase genes were involved in these activities.It shows the cuticular layer is formed at pupae,which as a permeable membrane involved in a large number of transport processes at moth.3.The KEGG enrichment analysis results showed that 11 pathways of the first 20 pathways with the least significant Q values were related to metabolism process.It can be thought that the mucous gland formation is an energy-consuming process,there are a large number of catalytic enzymes and glycoproteins in the metabolic process,which may provide a large amount of energy for the formation of mucus.4.The COG classification analysis identified 14 genes that fell into "Inorganic ion transport and metabolism".These genes might create the proper ionic environment in the CG for the mucous formation.Cu/Zn superoxide dismutase is a large family of cytosolic antioxidant proteins reducing oxidative stressed,it might help maintain homeostasis in vivo.5.We identified three candidate genes involved in the synthesis and secretion of mucinous glands:Serpin-28,GSTe3,AKR2E4-like,successfully cloned and expressed its target proteins,and prepared polyclonal antibodies.The expression pattern of target genes in the 3rd day 5th instar tissues showed that they were expressed in all tissues,but the expression levels were different.The relative expression level of the target gene in the different developmental stages of mucinous glands was consistent with the expression of protein levels,but the expression levels were also different.
