Screening Of Ng Correlative Genes Of Silkworm,Bombyx Mori Based On Transcriptome-Proteome Associative Analysis | Posted on:2019-12-13 | Degree:Master | Type:Thesis | Country:China | Candidate:L L Yang | Full Text:PDF | GTID:2493305495997459 | Subject:Biochemistry and Molecular Biology | Abstract/Summary: | PDF Full Text Request | Bombyx mori is a kind of model inset with import economic value and its collterial gland is the place for secretion and storage of mucus,which plays an important role in fixing and protecting the eggs.In this study,analysis of comparative transcriptome and proteome was carried out using the H9 and P50 strains of Bombyx mori and biological analysis of differentially expressed genes was investigated.The results are as follows:1.Comparative transcriptomics of the H9 and P50 collterial glandCollterial gland of maiden moth of H9 and P50 were used in comparative transcriptome sequencing.A total of 1509 DEGs were obtained for functional annotation,with 1001 DEGs were up-regulated(P50 >H9)and 508 DEGs down-regulated(P50 <H9).Differentially expressed genes are more related to the transport of amino acids,sugars and inorganic ions,mainly involving lipid metabolism and glucose metabolism.2.Comparative proteomics of the H9 and P50 collterial glandCollterial gland of maiden moth of H9 and P50 were used in comparative proteomics.A total of 4,423 proteins were identified,of which 633 proteins were up-regulated(P50>H9)and 814 proteins down-regulated(P50 <H9).Differentially expressed proteins are more involved in metabolic processes and catalytic activity,and are primarily involved in the processing of endoplasmic reticulum proteins.3.Crosstalk analysis between transcriptome and proteome of the H9 and P50 collterial glandA correlative analysis of proteome and transcriptome of the H9 and P50 collterial gland suggested that the expression of 115(Up-Up)of 633 up-regulated proteins were consistent with those in the transcriptome,and 113(Down-Down)of 814 down-regulated proteins were consistent with the transcriptome.4.Cloning and pattern expression of the differentially expressed genesFour correlative genes of GALT,NANA-Lyase,GALK-LX2 and GFPT1-Like that might be involved in the mucus synthesis and secretion of collterial gland,were initially identified,followed by cloning and analysis of their pattern expression.The target genes were expressed in all examined tissues in the 3rd day 5th instar of B.mori larvae,but the relative expression level in different tissues was different.There were also significant differences in the expression levels of different genes in H9 and P50,as well as the different development stage.By analyzing the transcriptome and proteome of the collterial gland of H9 and P50 and screening the correlative genes related to Ng traits,the basis for further study on the development and secretion of collterial gland were provided. | Keywords/Search Tags: | Bombyx mori, Collterial gland, Transcriptome, Proteome, Crosstalk analysis | PDF Full Text Request | Related items |
