| Tobacco incense-base modules are one of the indispensable additives,which play an important role in providing unique flavor of tobacco.The tobacco industry proposed“independent fragrance modulation” to replace imported incense-base modules with imitation incense to overcome the problem of “excessive dependence” on imported incense base modules.Therefore,it is necessary to establish a feasible method to evaluate the substitutability of tobacco incense-base modules,including consistency evaluation based on fingerprint and safety evaluation of products after replacement of incense-base modules.In order to overcome the limitation that one-dimensional fingerprint is not comprehensive and sensitive enough to evaluate the consistency of samples,it is considered an effective solution to establish multi-fingerprint to evaluate the consistency of samples.At the same time,the chemical composition of tobacco products will change by the addition of incense-base modules.So,evaluating the safety of tobacco products which added the imitation flavor sample is another important part of establishing substitutability evaluation method and guarantees the rights and interests of consumers.This study established the multu-dimensional fingerprint based on GC-Orbitrap MS,UV,and IR.And it formed a consistency evaluation method by combining with multivariate statistical analysis to evaluate imitation flavor.Then the safety of tobacco products which added imitation flavor were assessed by neutral red uptake(NRU)assay,in vitro micronucleus(IVMN)assay,and salmonella typhimurium reverse mutation(Ames)assay.Finally,a set of method and procedure for comprehensive evaluation of the substitutability of tobacco incense-base modules are established.The results were as follows:1.In order to construct a consistency evaluation method based on multi-fingerprint and more comprehensively characterize sample component information,we optimized the separation analysis method.Combined with Pearson correlation coefficient(PCCs),the multi-fingerprint was established based on gas chromatography-high resolution mass(GC-Orbitrap MS),ultraviolet spectroscopy(UV)and infrared spectroscopy(IR).The results showed that the chromatographic PCCs ≥ 0.8 and spectral PCCs ≥ 0.9between batches could be used to establish fingerprints.2.In order to build a consistency evaluation method,we further combined multi-fingerprint with multivariate statistical analysis to comprehensively evaluate the consistency between the imitation flavor samples and multi-fingerprint.Fingerprinting combined with principal component analysis(HCA)and hierarchical cluster analysis(PCA)based on GC-Orbitrap MS;UV fingerprint combined with linear regression analysis;IR fingerprint combined with second derivative analysis was used to determine the imitation flavor samples with high consistency.Finally,based on the three consistency evaluation results,the highest consistency of each tobacco incense-base modules is determined for the imitation flavors.3.In order to evaluate the safety,NRU,IVMN and Ames assay were used to evaluate the effects on nongenotoxic and genotoxic of tobacco products which added the imitation sample.The results showed that the nongenetic toxicity and mutagenic toxicity of tobacco products did not increase with the addition of each imitation sample.However,the addition of imitation samples could increase the genotoxicity of chromosome damage in tobacco products except FAJ-4.Therefore,it is necessary to control the degree of concentration in the use of imitation flavor,so as to guarantee the safety of the product and to protect the consumer’s interests.4.A set of substitutability evaluation methods and procedures for tobacco incense-base modules are formed by the combination of consistency evaluation and safety evaluation... |
