| Gastric motility disorder or deficiency is a manifestation of decreased peristaltic force of gastric muscle contraction,which affects people’s life and health.Studies have shown that the pathways affecting gastric motility are mainly divided into neuromodulation,humoral modulation and gastric muscle modulation.Currently,a variety of targeted drugs and interventions have been developed based on these three regulatory approaches,but drug therapy is usually accompanied by side effects.Dietary interventions are more acceptable than drugs and are more suitable for people with milder symptoms of insufficient gastric motility.Some medicinal foods such as hawthorn and Tangerine peel have been used to promote gastrointestinal digestion,but their effectiveness,mechanism of action and dosage to promote gastric motility are not yet clear.Therefore,This paper is based on the screening of medicationfood homologous foods with gastric motility effect.We investigated their mechanism of action and dose-effect relationship,obtained dietary formulas for improving gastric motility through different food combinations,and further investigated the synergistic effect of probiotic fermentation.The main findings were as follows:L-arginine(L-arg)was used to construct a rat model of gastric motility disorder,and chicory,Tangerine peel,chicken gizzards,hawthorn and phyllanthus emberium were used to investigate their in vivo pro-gastric motility efficacy and mechanism targets.The results showed that one week after gavage intervention in rats with gastric motility disorder,hawthorn,chicory and phyllanthus emberium significantly increased the effects of feeding,gastric emptying rate and intestinal propulsion rate,and chicory also significantly repaired as sinus secretory gland layer with epigastric motility effect;hawthorn significantly increased pepsin activity and serum hunger hormone(Ghrelin,GHRL)expression level;chicory significantly decreased total gastric acid concentration,free acid concentration and chicory significantly reduced total gastric acid concentration,free acid concentration and free acid excretion rate,and significantly increased serum GHRL expression level and gastric sinus c-kit m RNA transcript level;phyllanthus emberium significantly reduced gastric free acid concentration and excretion rate,and significantly increased serum Gastrin(GAS)level.These results suggest that hawthorn,chicory and yucca have good pro-gastric motility effects and different mechanisms.The literature suggests that the optimal dose of hawthorn for gastric motility is 10 g/d,while the optimal doses of chicory and yucca are unknown.Therefore,low,medium and high dose groups of chicory(4.5 g/d,9 g/d,18 g/d)and low,medium and high dose groups of Euphorbia(2.3 g/d,4.5 g/d,9 g/d)were set to investigate their pro-gastric motility effects.The results showed that chicory low and medium doses and phyllanthus emberium low dose significantly increased daily food intake and gastric emptying rate,chicory medium dose significantly increased intestinal propulsion rate,and each dose of chicory and phyllanthus emberium low dose significantly repaired the secretory glandular layer of gastric sinus with epigastric motility effect;chicory medium dose and phyllanthus emberium low,medium and high dose significantly decreased free acid concentration;chicory low and medium dose significantly increased serum GHRL expression level,and phyllanthus emberium low and medium dose significantly increased serum GAS expression level.The low,medium and high doses of chicory significantly increased the level of c-kit m RNA transcription and the medium dose significantly increased the level of SCF m RNA transcription,and the low,medium and high doses of chicory and the low dose of Eugenia significantly increased the percentage of whole mucosa in the secretory gland layer of gastric sinus.The best starting dose of chicory was determined to be 9 g/d for the medium dose and 2.3 g/d for the low dose.The pro-gastric motility effects of various formulations were investigated by combining two or all of Hawthorn,Chicory and phyllanthus emberium.The results showed that all the formulations significantly enhanced the intake,gastric emptying rate and intestinal propulsion rate,and the pro-gastric motility effect was significant.Among them,the combination of hawthorn(10 g/d),chicory(9 g/d)and phyllanthus emberium(2.3 g/d)showed the best effect in gastric emptying rate,intestinal propulsion rate,whole mucosal percentage of the secretory gland layer of the gastric sinus,protease activity and expulsion rate,serum GHRL,GAS and MTL expression levels,and c-kit/SCF m RNA transcript levels,and was therefore determined as the best dietary The best dietary formula was determined.The optimal dietary formula was fermented with probiotics and applied to human gastric adenocarcinoma cells AGS to investigate the feasibility of fermentation potentiation.The results showed that the addition of 8% of Streptococcus thermophilus M2 fermentation supernatant to the cell culture medium significantly increased the release of GAS from AGS cells,and the addition of 7% of Streptococcus thermophilus M2 fermentation solution significantly increased the transcription level of GHS-R m RNA,and both effects were significantly better than the addition of the same concentration of the original dietary formula.This indicates the feasibility of probiotic fermented dietary formulations to enhance the progastric motility effect. |
