Traditional Chinese medicine sachet is a kind of perfume sachet made of fragrant Chinese herbs.It has the effect of fragrance to avoid distemper and prevent disease.It is commonly used in folk to refresh,prevent mosquitoes and treat influenza.There are many kinds of Chinese herbal sachets,commonly used are Guanghuoxiang,Aiye,Shannai,Cangzhu,Baizhi,Dingxiang and so on.The production method is simple.The Chinese medicinal materials need to be washed,dried,crushed,then put into a silk bag and stitched to seal.Wearing traditional Chinese medicine sachets can form a high concentration of small environment around the mouth and nose,enhancing resistance and immunity.The novel coronavirus pneumonia is an infectious respiratory disease caused by the SARS-Co V-2 virus,also known as COVID-19.The virus first emerged in late 2019 and quickly spread to many countries and regions around the world,causing serious impact.The main route of transmission is through respiratory droplets and close contact.Common symptoms include fever,coughing and difficulty breathing.There are no specific drugs or effective preventive vaccinations,but there are steps that can be taken to reduce the risk and severity of infection.In the previous study,the experimental study on the novel coronavirus pneumonia by extracting three drugs of Cangzhu,Aiye,Guanghuoxiang from sachets confirmed that Cangzhu,Aiye,Guanghuoxiang has the effect of clearing heat,anti-inflammation and anti-novel coronavirus pneumonia,but the specific mechanism of action of Zisuzi,Bohe on novel coronavirus pneumonia is still unclear.On the basis of the above studies,based on network pharmacology and molecular docking,this paper carried out a study on the mechanism of action of the effective ingredients in sachet against the novel coronavirus pneumonia,providing reference and scientific basis for the research and development of innovative sachet drugs.Research contents of this paper :(1)In this paper,the pharmaceutical ingredients in the sachets were used as research materials.Three drugs were selected in the sachets in the early stage,including Cangzhu,Aiye,Guanghuoxiang.Three drugs were searched and screened through TCMSP database to obtain effective active ingredients,including wogonin,quercetin,genkwanin,etc.The active ingredients were combined with Gene Cards database to screen 24 key targets for the treatment of COVID-19,among which the core targets included IL6,CXCL8 and VEGFA.GO enrichment analysis showed 578 biological processes,11 cellular components and 39 molecular functions,and KEGG Pathway enrichment analysis showed 150 pathways.The PPI network was constructed to explain how the three active ingredients of perfume sachets function.(2)Theoretical verification of network pharmacology using molecular docking.Three active components of wogonin,quercetin,genkwanin were screened for molecular docking with three target proteins IL6,CXCL8 and VEGFA.It was found that the binding energy of both baicalin quercetin genkwanin and IL6 CXCL8 VEGFA core target was less than-5.0kcal/mol.These results suggest that the active components of Cangzhu,Aiye,Guanghuoxiang in sachets can be used to regulate the target for COVID-19 treatment.(3)In order to further study the mechanism of sachet against COVID-19,two other drugs were added on the basis of previous experiments: Perilla seed and mint.Through network pharmacology and the study of five components in sachet,27 target sites were obtained.The enrichment analysis of the newly added core target ALB GO was basically consistent with the enrichment Pathway of KEGG Pathway and the previous experiment.The binding energy of the newly added active components luteolin and naringin in the molecular docking results with the core target was also less than-5.0kcal/mol,stable binding activity.
