Traditional Chinese medicine(TCM)is a mixture of multiple chemical components,and its effectiveness and specificity are difficult to be determined by one or a few components.The holistic concept in the traditional philosophy of TCM also guides TCM to be used as a whole and doesn’t focus specifically on a single drug or a single ingredient.In addition,the different processes of harvesting,processing,and pharmaceuticals of similar herbs lead to different contents of active ingredients,the use of plants near the edge of the medicine to disguise the real,substandard for superior phenomenon also prevails,these factors greatly limit the modernization and to the international market of TCM.In this thesis,an oscillating system of H2SO4-Ce(SO4)2-KBrO3-CH2(COOH)2(HOOCC(H)OH)2 double organic substrate was established.Taking this system as a blank system,adding TCM.Platinum electrode was used as indicator electrode,saturated calomel-electrode as reference electrode,and electrochemical workstation was used to detect its electrochemical fingerprint.The cluster analysis,discriminant analysis by SPSS software and similarity analysis of the electrochemical fingerprint were used to identify the origin of Astragalus and Radix Polygalae.Specific research contents and conclusions are as follows:1.Specificity of electrochemical fingerprint of TCM.Using the above blank system as the oscillation system,the electrochemical fingerprints of different kinds of TCM as well as the same kind but with different origins,different preparation methods,different slicing methods,different addition amounts,different parts and different growth years were examined and analyzed.It was found that the shape,trend and the size of each characteristic parameter of the different TCM were obviously different.Therefore,the electrochemical fingerprint database of different TCM could be established based on this method.When it is necessary to distinguish the unknown sample,the electrochemical fingerprint of it can be determined and compared with the fingerprints in database.2.Verification of reproducibility of electrochemical fingerprint of TCM.Using the above blank system as the oscillation system,the electrochemical fingerprints of different TCM or the same TCM with different additions,different preparation methods and different origins of the powder were repeatedly detected and analyzed to discover the characteristic parameters and verify the reproducibility,and it was found that the method was better reproducible for detecting the electrochemical fingerprints of TCM and could be used for the subsequent identification of the origin and quality of TCM.3.Origin tracing of Radix Astragali based on electrochemical fingerprint.In the above blank system,Radix Astragali of different quality and origin were added respectively.The electrochemical fingerprints of Radix Astragali were measured.The characteristic parameters were analyzed and function fitting was performed with the additive amount of Radix Astragali.Finally,the maximum amplitude and maximum potential of the seven characteristic parameters were selected.Because they have a good linear relationship with the addition amount of Radix Astragali.The cluster analysis and discriminant analysis were carried out by SPSS software.The results showed that Radix Astragali located in the Qinling Mountains of central China and other producing areas were obviously divided into two categories.Since the nature and content of chemical components of TCM are mainly related to natural conditions such as climate,rainfall,altitude and sunshine time in the locality.And the result of traceability analysis is basically consistent with the classification of real origin,it can be considered that SPSS analysis combined with electrochemical fingerprint can be used to trace the origin of Radix Astragali.4.Identification of Polygala Tenuifolia origin based on electrochemical fingerprint.In the above blank system,the Polygala Tenuifolia of different quality and different origin were added respectively,and their electrochemical fingerprints were measured.The characteristic parameters were analyzed and linear fitting was performed with the addition amount of Polygala Tenuifolia.Finally,the induction period and maximum potential of the seven characteristic parameters were selected.Because they have a good linear relationship with the addition amount of Polygala Tenuifolia.Based on the similarity analysis,the similarity between other origin and Shanxi Province was analyzed.The results showed that the method was consistent with the geographical classification of the origin of the Polygala Tenuifolia.It was considered that the method could be used to identify the origin of the Polygala Tenuifolia.Compared with other methods for identifying the origin of TCM,this method does not require complex pre-treatment of TCM samples,can retain the cluster information of the samples to the maximum extent,and detect the characteristics of TCM as a whole,which is in line with the traditional "holistic" treatment concept of TCM.Moreover,the method has the merit of strong characterization,high sensitivity,good stability,simple operation and low analytical cost,which is suitable for further popularization and has good application prospects.
