Dictyophora rubovovatar belonging to the family of Hallaceae,is a valuable resource with abundant active functions such as blood pressure lowering,anti-inflammatory,and immune regulatory effects.The fruiting body of D.rubrovalvatar is composed of a pileus,stipe and volva.The pileus and volva are usually discarded when consumed,leading to a significant waste of this valuable resource.Therefore,it is of great economic value and social benefit to study the endogenous components and functions of the pileus and volva of D.rubrovalvatar for efficient utilization of this resource.There are few reports on the analysis of the active components and antioxidant activity of D.rubovolvatar.This study focused on the research object of D.rubovolvatar.Firstly,we prepared ethanol extracts of different parts of D.rubovolvatar(pileus,stipe,volva)and analyzed their components and antioxidant properties.Subsequently,response surface methodology was used to optimize the extraction process of polysaccharides from D.rubovolvatar,and the physicochemical properties and antioxidant properties of the extracted polysaccharides were studied.(1)We prepared alcohol extracts from various parts of D.rubovolvata using an ultrasonic extraction method and 80%ethanol as solvent.Composition determination findings were found that the extracts had high protein and total polysaccharide contents.The stipe extract had the highest protein content of 83.08±1.22 mg/g.The total polysaccharide and polyphenol contents of the pileus and volva extracts were higher than those of the stipe extract.The pileus extract had a polysaccharide content of 53.66±1.43 mg GE/g and a polyphenol content of 7.02±0.27μg GAE/g,while the volva extract had a polysaccharide content of 51.30±1.41 mg GE/g and a polyphenol content of 6.20±0.16μg GAE/g.The stipe extract had a polysaccharide content of 49.74±1.73 mg GE/g and a polyphenol content of 5.60±0.09μg GAE/g.(2)In vitro antioxidant experiments(DPPH,ABTS,FRAP)showed that the ethanol extracts from the pileus,stipe,volva all exhibited good antioxidant activity.The ability of pileus and stipe to clear DPPH and reduce iron ions is better than that of volva(Pileus:DPPH:IC50=459.1±4.51μg/m L,FRAP:IC50=370.6±8.72μg/m L,Volva:DPPH:IC50=681.6±31.94μg/m L,FRAP:IC50=380.8±12.26μg/m L.,Stipe DPPH:IC50=710.8±14.93μg/m L,FRAP:IC50=546.5±3.25μg/m L).By establishing cellular oxidative stress model,the ethanol extract from D.rubovolvatar(200μg/m L)could significantly reduce ROS production,LDH secretion,and MDA content in H2O2-induced Ha Ca T cells,and significantly increase SOD activity.In addition,the ethanol extracts of all three parts upregulated the expression levels of NQO1 and HO-1 proteins through the Nrf2 pathway,and the upregulation effect of the volva was the most significant.The aboved result revealed that the ethanol extracts of all parts of D.rubovolvatar had antioxidant effects,compared to pileus and stipe,the volva showed superior antioxidant properties.(3)The extraction conditions of polysaccharides from D.rubrovalvata were optimized through single-factor and response surface experiments.The optimal extraction conditions were determined as follows:solid-liquid ratio of 1:25(g/m L),extraction time of 3 hours,extraction temperature of 80°C,and extraction repeated twice.Under these conditions,the extraction rate of polysaccharides was(9.68±0.08)%.The extracted polysaccharides were then subjected to protein removal using the Sevage method and decolorization using macroporous resin,resulting in purified D.rubrovalvata polysaccharides(Drsp).UV-vis absorption spectroscopy confirmed the purity of the extracted Drsp,which was free from pigments and nucleic acids and contained little or no protein.FTIR spectroscopy indicated that Drsp was a pyran-type polysaccharide containingβ-glycosidic bonds.Ion Chromatography determined the monosaccharide composition of Drsp as mannitol(1.65%),rhamnose(6.98%),glucose(80.32%),xylose(0.38%),and galactose(10.67%).(4)Research on the antioxidant properties of polysaccharides from D.rubrovalvata indicated that Drsp could effectively eliminate ABTS radicals and reduce iron ions(ABTS:IC50=325.15±5.5μg/m L;FRAP:IC50=613.1±39.1μg/m L).In addition,Drsp could alleviate oxidative stress damage in Ha Ca T cells induced by H2O2.The cell scratch experiment results showed that Drsp could enhance the migration ability of Ha Ca T cells and promote wound healing.This study revealed that the extracts of D.rubovolvatar have good antioxidant properties,and the antioxidant properties of the pileus and ethanol extracts are superior to those of the stipe extract.This research provided a theoretical basis and experimental support for the full utilization and development of the resource of D.rubovolvatar.Figure[30]Table[29]Reference[132]... |
