Objective A total nutritional formula for inflammatory bowel disease supplemented with Lactobacillus plantarum GL-5 was innovatively developed,and it was adjunct applied to mice with ulcerative colitis induced by sodium glucan sulfate to observe its efficacy,evaluate its safety,and explore its therapeutic mechanism.Methods 78 C57BL/6 female mice were randomly divided into 6 groups: normal group,model group,self-made formula group,ansu group,mesalazine group,and combined treatment group.After 7 days of adaptation,except for group N,the mice were given purified water,and the other groups were given 3.5%DSS aqueous solution for 7 days.The body weight,food intake,water intake,and adverse events of the mice were collected daily during the whole experiment.After the intervention,eyeballs were collected and blood samples were collected.Physiological and biochemical indexes such as liver function and kidney function were measured in serum and combined with general physiological conditions during the experiment to evaluate its safety.At the end of the modeling and the end of the intervention,the disease activity index was scored twice to evaluate the clinical effect.Clinical efficacy was evaluated by two disease activity index scores and proximal colon histopathological scores before and after the intervention.The therapeutic mechanism was explored by intestinal flora measurement(α-diversity,β-diversity,etc.)and non-targeted metabolomics analysis(differential metabolites and metabolic pathways).Results The diet and water intake of the GL-5 formulation group and combined treatment group were significantly recovered.The weight recovery rate and final body weight of mice in the GL-5 formulation group,ansu group,and combined treatment group were better than those in the mesalazine group and model group(P<0.05).The incidence of adverse events in both groups was lower than that in the model group,and compared with the normal group,there was no statistical significance in the physiological and biochemical indexes of the GL-5 formulation group(P>0.05).After the intervention,the disease activity index of each group was lower than that in the modeling stage.Compared with the model group,the scores of the GL-5 formulation group,ansu group,and combined treatment group were lower,with statistical significance(P<0.05).The proximal colon section and pathological evaluation of the GL-5 formulation group and combined treatment group showed that the intestinal mucosal structure was healthier and more recovered(P<0.05).Compared with the model group,the abundance of beneficial bacteria increased in the GL-5 formulation group and combined treatment group,while the abundance of harmful bacteria decreased,and the diversity of intestinal flora was richer.Compared with the mesalazine group,the GL-5 formulation group showed improved inflammation reduction and expression of metabolic small molecules.Conclusion Adjuvant treatment of UC mice with modified and added GL-5 full nutritional formula food has guaranteed safety and significantly improved UC symptoms.The GL-5 formula group showed efficacy no weaker than the combined treatment group and mesalazine group,and its mechanism may be related to increasing intestinal flora diversity,inhibiting inflammation,and promoting the expression of small molecules that are beneficial to nutritional metabolism.
