| "Double protein" refers to the combination of plant protein and animal protein to achieve accurate interaction between the two proteins,which plays an important supporting role in regulating and controlling diet structure.Soybean protein is the most commonly used plant protein.Besides soybean,there are many beans with high nutritional value.Combined with the previous research results of our research group,black bean protein except soybean protein was selected as the research object in this study,and the soy protein and whey protein were mixed in a ratio of 1:2.Then,the influence of legume-whey "double protein" diet on the immunity of rats was explored by measuring immunological indicators at the in vivo and cellular levels.The 3-week-old male SD rats were fed with casein(CP),soybean protein(GM),black bean protein(BSP),whey protein(WP),black bean-whey “double protein”(BS-WP)and soybean-whey “double protein”(GM-WP)diets.After 8 weeks of feeding,The immunoglobulin,serum biochemical indexes,immune organ index,splenic lymphocyte proliferation index,natural killer(NK)cell activity,macrophage phagocytosis index,cytokine release and other indexes were determined.The results are as follows:(1)The amino acids in the proteins of each group were analyzed by hydrolysis method.The AAS values of amino acids in the BS-WP and GM-WP groups were both greater than or close to 1,indicating that the amino acid composition of each group was reasonable.(2)For serum biochemical indexes,compared with CP group,serum protein(TP)content of rats in GM-WP group was increased by 49.06±1.94 g/L,total cholesterol was decreased by 2.86±0.59mmol/L and triglyceride was decreased by 1.14±0.13 mmol/L.Serum protein content of BS-WP group increased by 47.85±1.94 g/L,total cholesterol decreased by 2.66±0.54 mmol/L,triglyceride decreased by 1.23±0.21 mmol/L,serum urea nitrogen content of GM-WP group was significantly increased(P<0.05),BS-WP group had no significant difference(P≥0.05).For the immunoglobulin(Ig A,Ig G,Ig M),there was no significant difference between the single protein groups(P≥0.05),the immunoglobulin content of rats in the “double protein” group was significantly increased(P<0.05),and the Ig G content of the BS-WP group was higher than that of the GM-WP group.(3)Compared with the single protein group,the immune organ index and NK cell activity of rats in the “double protein” group were significantly increased(P<0.05).(4)“double protein” histone digestion increased the phagocytosis index of macrophages,promoted the secretion of IL-2 and IFN-γ in spleen lymphocytes,and inhibited the secretion of IL-4.Diproteomic digestives promoted the secretion of IL-1β,IL-6,TNF-α,i NOS and LZM in macrophages,and the effect was most obvious when the concentration of protein digestives was 100μg/m L.In conclusion,the legume-whey “double protein” diet can regulate the immune condition of the body and play an important role in maintaining the health of the body,and the immune promotion effect of the BS-WP group is better than that of the GM-WP group.The results of this experiment can provide a theoretical basis for the development of legume-whey “double protein” products. |
