Purpose:The micro nano topological structure modified coating containing Mg2+/Cu2+was constructed on the surface of pure titanium by alkali heat method and impregnation method.The characterization of the coating were detected by material science method,the synthesis of the material was verified,and the antibacterial ability of the coating on two kinds of bacteria and its effect MC3T3-E1 and human umbilical vein endothelial cells(HUVECs)were discussed.Method:1.The micro nano topological coatings containing Mg2+,Cu2+and Mg2+/Cu2+were prepared on the surface of titanium substrate.ICP test results show that the ionic dissolution rate and the amount of ions on the coating surface decrease gradually with time,and the bimetallic ion group has a certain slow-release effect.2.The surface morphology,element composition and content,phase composition,element valence and ion dissolution rate of the coating were detected by scanning electron microscope,energy spectrum analyzer,X-ray diffractometer,photoelectron spectrometer and inductively coupled plasma mass spectrometer.3.Plate counting method was used to detect the antibacterial ability of different coatings against S.aureus and E.coli.4.The effect of the modified coating on the proliferation and differentiation of osteoblasts was evaluated by detecting the proliferation of osteoblasts,the ability of protein adsorption,the activity of alkaline phosphatase(ALP)and the expression level of osteogenic related genes.5.The ability of promoting angiogenesis was evaluated by detecting the effects of each group of samples on the proliferation and migration of endothelial cells,the secretion of vascular endothelial growth factor(VEGF)and the expression level of angiogenesis related genes.Result:1.The micro nano topological coatings containing single metal ions Mg2+,Cu2+and bimetallic ions Mg2+/Cu2+were successfully prepared on the surface of titanium substrate by alkali thermal method and impregnation method.ICP test results showed that the ion dissolution rate and ion dissolution amount on the coating surface decreased gradually with the passage of time.2.The modified coating has a good inhibitory effect on E.coli and S.aureus,and the coating containing bimetallic ions Mg2+/Cu2+has the most prominent antibacterial ability.3.The modified coating can improve the proliferation of osteoblasts,the ability of protein adsorption,the activity of alkaline phosphatase and the expression level of osteogenic related genes,among which the bimetallic ion group is the most prominent.4.The modified coating,especially the group containing bimetallic ions,can promote the proliferation and migration of endothelial cells,the secretion of vascular endothelial growth factor and the expression of angiogenesis related genes.Conclusion:1.The micro nano topological structure,micro nano topological structure coating containing single metal ions Mg2+,Cu2+and micro nano topological structure coating containing bimetallic ions Mg2+/Cu2+were successfully constructed on the surface of pure titanium by alkali thermal method and impregnation method.The bimetallic ion group has a certain slow-release effect.2.The Mg2+/Cu2+micro nano topological structure coating containing bimetallic ions has excellent antibacterial ability,and can promote the proliferation and differentiation of osteoblasts and endothelial cells.
