| Oatsβ-glucan(OG)has a simple sugar unit structure,linear and hydrophilic groups,and is easily crosslinked with polyphenols to enhance its antioxidant activity.Quercetin(QE)has been widely used as a natural antioxidant in the food field,but its bioavailability is low due to its low solubility and easy loss during extraction.As one of the main components of human food,sugar and phenol easily form complex during digestive process.However,there is relatively little research on the formation mechanism,digestive characteristics and biological activity of sugar phenol complex.The crosslinking of glycophenols into complex can reduce the degradation of polyphenols in the intestinal environment,improve the infiltration of polyphenols in the small intestine,and even increase the content of polyphenols in the blood.This research adopts the radical catalyst,hydrogen bonding adsorption technology,respectively,the preparation of oat beta glucan and quercetin covalent(COGQ)and non covalent compounds(NOGQ),using the gel exclusion chromatography,infrared spectrum and nuclear magnetic spectrum method for determining its structural features,in Caenorhabditis elegans(C.elegans)as model,the complex anti-aging activity.Gel exclusion chromatography showed that the relative molecular weight of NOGQ and COGQ were 354 k Da and 349 k Da,respectively,with little difference between them.The infrared spectra showed that the new infrared absorption peaks at 1660,1562 and 1243 cm-1were amorphous and amorphous.NMR spectra show that the complex can generate three new proton signals belonging to phenyl protons and three monosaccharide residues in the range of chemical shift 6.5~7.5.DPPH,ABTS,reducing power,hydroxyl radical and superoxide radical scavenging capacity of the compounds were studied respectively.The results showed that the free radical scavenging rates of NOGQ and COGQ were higher than OG at the same concentration,indicating that the compound had strong antioxidant and free radical scavenging ability.Compounds of anti-aging activity study showed that compared with the blank group,COGQ and NOGQ can significantly increase the average life expectancy of Caenorhabditis elegans(P<0.05),and two groups of SOD and CAT activity were significantly increased(P<0.01),two kinds of compounds may by increasing the antioxidant capacity of the body,and exert its efficacy of life extension.The results provide a reference for the synthesis of glycophenol complex and its application as a new dietary fiber in anti-aging functional food. |
