There are about 250 species of Valerians officinalis all over the world,most of which are distributed in temperate regions.There are 28 species and 2 varieties of valerians plants in China,mainly produced in the southwest,northwest and northeast.The modern pharmacological and clinical trials showed the volatile oil extracted from Valerians Herb had activity such as sedation,hypnosis,antisepsis,antivirosis,protecting heart,anticancer and so on.In this paper,the extraction process,chemical composition and quality control methods of volatile oil from Valeriana officinalis L.Var.1atifolia Miq were widely studied in Guizhou.The main conclusions are as follows:The optimum extraction process of volatile oil from roots of Valeriana officinalis in Guizhou province was studied and its chemical composition analyzed.Volatile oil from roots of V.officinalis was extracted by steam distillation method,distillation time,solid-liquid ratio,soaking time,degree of smash and other factors were investigated with extraction ratio of volatile oil as index,extraction technology of volatile oil from roots of V.officinalis was optimized by single test and response surface methodology.The optimum conditions of steam distillation were as follows:grinding degree 45 mesh,solid-liquid ratio 1:10.3,soaking time 2.3 h;Under these optimized extraction conditions,yield of volatile oil was 1.59%.Molecular distillation was used for the further purification of the volatile oil from roots of V.officinalis.Adopt gas chromatography to determine the Content of Valeriana volatile oil which was further purified by Molecular distillation equipment.The best process parameters,including temperature,feed rate,and scraping rate of molecular distillation technique for separating were investigated with the content of bornyl acetate as index,the volatile oil were studied by response surface methodology based on single factor experiment.The optimal extraction processing parameters were material-feeding rate of 82 m L/h,distillation temperature of 68? and scraping rate of 188 r/min.Under the optimal processing conditions,the quality of volatile oil was markedly improved,and the content of bornyl acetate was 77.46%.A quality control system of the volatile oil of V.officinalis was established,lay a foundation for new agent research and development.Results:The GC methods of quantitative quality control and specific chromatograms for the volatile oil were established.The sum of the specific peaks area in feature chromatograms and mutual mode of eleven batches volatile oil more than 0.9 of the total peaks area,the average percentage was 91.36%.The effective components from valeriana volatile oil was analyzed by gas chromatography-mass spectrometry(GC-MS).Results showed that there were 27 kinds of chemical composition were identified from volatile oil,the main components of which were sesquiterpene and monoterpenes and it contained olefin,alcohol,ketone,ester,ether compounds,respectively.Bornyl acetate and camphene took dominant position,their relative mass fraction were 50.69%,22.93%.
