| A small tree of the genus Buckthorn in the family Molagniae,buckthorn has a number of pharmacological effects,including high flavonoid content.A small tree of the genus Buckthorn in the family Molagniae,buckthorn has a number of pharmacological effects,including high flavonoid content.Despite these potential medicinal properties,little research has been done on seabuckthorn.Therefore,this paper discussed the effect of deep processing of sea buckthorn.By obtaining high value-added flavonoids,the waste problem of sea buckthorn residue was successfully solved,and good economic benefits were created for enterprises.The main raw material is fruit juice residue,from which total flavonoids are extracted and separated.The conventional solvent extraction method and ultrasonic assisted extraction method were used to compare the extraction of active components from seabuckthorn fruit residue.The contents of total flavonoids in Seabuckthorn waste residue were compared by using ethanol as solvent combined with traditional solvent and ultrasonic method.Through orthogonal experiment and single factor experiment,the best process parameters were obtained: 60% ethanol,70℃,1:40 solid-liquid ratio,2 hours extraction;Ultrasonic assisted extraction technology was adopted,with 50% ethanol,1:25 solid-liquid ratio,20 minutes and 0.4 k W ultrasonic power as the optimal process parameters.It was found that the optimal extraction parameters of ultrasonic assisted extraction were 20 minutes,and the total flavonoids yield reached 6.45%.Compared with traditional extraction methods,ultrasonic assisted extraction can significantly reduce the amount of solvent and extraction time.Flavonoids in seabuckthorn were analyzed by HPLC,including catechin,rutin,quercetin and isorhamnetin.The contents of catechin,rutin,quercetin,isorhamnosin and other components in the ethanol extract were the highest,followed by the solvent recovery through rotary evaporation,and the low content was the lyophilized powder solution after freeze drying.Therefore,in this study,sea-buckthorn bark was used as raw material to prepare sustained release tablets with microporous pore structure,and the formulation was optimized by Box-Behnken effect surface method to obtain the optimal formulation,and its sustained release effect was verified in vitro simulation.The study on the preparation of a solid disperse microporous osmotic pump tablets of total flavonoids of sea Buckthorn was initiated by a single factor experiment based on the cumulative dissolution degree of total flavonoids of sea buckthorn as an evaluation index,the integrative evaluation of drug release was carried out through the method of similar factors like Cosmos 2,where the prescription and technological parameters of this preparation were investigated.Subsequently,the optimized prescription was verified,and the results showed that the reproducibility between batches was good,and the zero-order drug release model fit was satisfied. |
