based on trying a new preparation method of blood viscosity quality control material.From the perspective of red blood cell morphology,particle size,deformation properties and macrorheological properties of whole blood,chitosan hydrogel microspheres are suspended The fluid form mimics the rheological properties of blood.(1)In the experiment,chitosan hydrogel was prepared by emulsification and crosslinking method.The oil-water ratio,stirring speed and mass fraction of chitosan in the preparation parameters were discussed,and a preparation method with a particle size close to red blood cells was obtained.In this experiment,when other conditions are the same,the oil-water ratio is 8:1,the stirring speed is 500(rad/min),and the mass fraction of chitosan is 3%,the particle size of the prepared chitosan hydrogel microspheres is more in line with the erythrocyte particle size.diameter requirements.(2)Use a cell deformer to measure the deformability of red blood cells and chitosan hydrogel microspheres,optimize the preparation process of chitosan microspheres prepared by emulsification cross-linking method,adjust the concentration of chitosan,the concentration of cross-linking agent glutaraldehyde,and the suspension The p H value changes the deformability of chitosan hydrogel microspheres.When the concentration of chitosan is 2.8%,the concentration of cross-linking agent experiment: the ratio of oil to water is 8:1,the rotation speed is 500(rad/min),the mass fraction of chitosan is 2.8%,and the concentration of cross-linking agent glutaraldehyde is 2%.The microspheres obtained by the preparation method were suspended in a 0.9wt%sodium chloride solution with a p H value of 5.0,which was recorded as the sample suspension.(3)The chitosan hydrogel microsphere suspensions with different dilution ratios were tested on two different viscometers,the capillary viscometer FASCO-3010 and the rotary rheometer MCR-102,and the same dilution ratio was compared with the whole blood viscosity.The detection rules were compared,and the relationship between the rheological properties of the chitosan hydrogel microsphere suspension and the rheological properties of blood was reflected from the perspective of the viscosity-shear rate relationship diagram.The experimental results show that the sample suspensions with different dilution ratios show non-Newtonian characteristics of shear thinning in the detection of different principles.It shows that the rheological properties of the prepared chitosan hydrogel microspheres suspension can reflect the rheological properties of whole blood to a certain extent.(4)In this experiment,two blood viscometers with different principles were used:capillary viscometer FASCO-3010 and rotary rheometer MCR-102 to detect chitosan hydrogel microsphere suspension samples.Once a month,a total of six times the viscosity indoor storage time stability test.According to statistics,the coefficients of variation of the viscosity values of the two instruments are 2.7%-5.2% and 5.5%-22.2%,respectively.The sample suspension showed certain stability in the detection of two different viscometers.The rheological properties of the prepared chitosan hydrogel microsphere suspension are close to the non-Newtonian properties of whole blood both microscopically and macroscopically.The study of chitosan hydrogel
