Carboxymethyl Chitosan (CMCS) is prepared by chitosan and monochloroacetic acid at alk a'lify and heat. It shows good water solubility at 4h reaction time and 50 癈 reaction temperature. The production is also analyzed by IR.CMCS's apparent viscosity is rising with thicker concentration and less temperature. CMCS's apparent viscosity has linear with the moment of NaCl. CMCS's apparent viscosity is also rising with adding NaCK Na2SO4 and NasPO to solution and it is also reducing with adding multivalent metallic salt -, sodium citrate and sodium tartrate. CMCS's apparent viscosity reach the max point at pH=10. CMCS is a kind of pseudoplastic fluid and it shows positive thixotropy but it shows negative thixotropy when add NaCK positive ion surfactant and sodium citrate.Glutraraldehyde(GA), epichlorohydrin(ECH), toluene diisocyanate(TDl) and hexamenthylene diisocyanate(HDI) are applied to crosslink CMCS respectively to get four aquagel: CMCS-GA , CMCS-ECH > CMCS-TDI and CMCS-HDI. The CMCS-GA swelling rate(SR) reaches 11.23 and the percent recovery reaches 101.1% when the proportion of GA and CMCS is 0.2:1. The CMCS-GA is a kind of sensitive gel to acid and temperature, the SR rises quickly from 5.09 to 11.3 when pH change from 1.0 to 7.0, while the SR reduce with temperature cut down. The CMCS-ECH's SR reaches 8.2 and the percent recovery reaches 113.6% when the proportion of ECH and CMCS is 0.8:1. The CMCS-ECH is a kind of sensitive gel to alkali and temperature. The SR rises quickly from 3.64 to 9.78 when pH change from 7.0 to 13.0, while the SR reach the min-point at 45and some NaCl can reduce the CMCS-ECH's SR. The CMCS-TDI and CMCS-HDI's swelling rate(SR) reaches 6.38 and 7.27 when the proportion of TDI and CMCS is 0.4:1 and the proportion of HDI and CMCS is 0.2:1. The CMCS-TDI and CMCS-HDI are also a kind of sensitive gel to acid, these above four gel are also analyzed by IR.The optimum condition of applied GA to crosslink CMCS to prepare themicrosphere is established: the concentration of CMCS is 3.0%, GA:CMCS=0.4:1, the reaction medium is atoleine, the volume ratio of atoleine and CMCS's solution is 3:1. the reaction time is 4h and reaction temperature at ambient temperature. To immobilize the Asl.398 neutral protease use above microsphere and get the optimum condition: GA:CMCS is 0.4:1 and enzyme:CMCS is 0.125:1. the reaction temperature is ambient temperature and the reaction time is 4h. The optimum temperature and pH of immobilized enzyme is 65 癈 and 6.0, the immobilized enzyme can protect a week at 5癈. the Km of immobilized enzyme and free enzyme is 1.04g/L and 5.66g/L. The CMCS-film has good penetrability. It can pass all kind of positive ion and negative ion. To all positive ion ,the osmotic coefficient's order is I > II >III, to monoacidic positive ion, the osmotic coefficient's order is KCl>NaCl>KBr>Kl>KF, to bivalence positive ion, the osmotic coefficient's order is ZnCl2>MgCl2>CaCl2> BaCb- The sustained-release drug microsphere of Hydrocortisone are prepared by applying hydrogel of ECH crosslinked CMCS. The preparation processes are optimized and the patterns of drug release are evaluated. The results have shown that: The drug microsphere with reasonable drug entrapment ratio and release velocity will be obtained if the amount of crosslinking agent and drugs is controlled successfully. The Hydrocortisone microsphere has a suitable release rate in artificial intestinal juice but a very low release rate in artificial succus gastrius, the cumulative release percentage after lOh is 88.9% and 43.5% respectively, the release patterns in the two mediums both fit the zero-order kinetics.Chitosan carboxylmethyl chitosan Aluminium carboxylmethyl chitosan and Magnesiumc arboxylmethyl chitosan were prepared. The six compounds were also investigated the determination of gastric acid neutralization in 37.5癈 water. The result was Ig products could neutralize 15.44mL, 30.79mL^ 41.75mL^ 69.85mLx 109.59mL and 67.86mL of 0.1172mol/L HC1 solution.
