The efficient delivery of exogenous drugs into the human body is an important component of biomedicine science.Conventional drug delivery relies on oral uptake and injection.Injection can release the drug rapidly and accurately,but it can damage the tissue and induce pain.Oral uptake reduces the efficient utilization of the drug,due to the first-pass effect.Drugs’ effected time is greatly prolonged by gastrointestinal tract digesting.Currently,the research focus of drug delivery is how to achieve more efficient,painless,controllable drug delivery.In this work,we reported a new transdermal drug delivery approach using the GHz hypersound system.The attenuation of gigahertz acoustic waves generated by the micro nano technology fabricated bulk acoustic resonator can produce rapid upward liquid motion,resulting in a needle-shaped jet flow assisting the permeation of drug molecules.Integrate the portable multi-channel signal source with the resonator to improve the efficiency and portability of the system.This research system has potential to develop the study of controllable,portable transdermal drug delivery.The system is expected to apply in commercial use.The main contents and the result of the research are as follows:1.Study the working mechanism of the hypersound resonator.Analyze the liquid needle’s mechanism(k Pa pressure)and working range(1000μm height),from the perspective of theory and experiment,by two-dimensional finite element simulation,microscope observation and the high-precision force sensor.2.Use isolated skins transdermal drug delivery experiments to explore the experimental parameters and methods of transdermal drug delivery based on the GHz hypersound system.Use 1 W,2.56 GHz(Cycle 1s,duty cycle 0.5)stimulation and improved skin preparation and anesthesia methods to conduct experiments.3.Based on in vitro transdermal drug delivery,carry out and improve the in vivo transdermal drug delivery experiment to increase reproducibility,and analyze the effect of drug delivery qualitatively and quantitatively.The transdermal drug delivery efficiency in the article is 3 % of the injection efficiency,while only causing minor damage to the skin surface,and the administration is controllable and stable.4.In the in vitro and in vivo experiment,the arrayed resonators are used to improve the drug delivery effect and compared with the drug delivery effect of the single resonator.In the thesis,the three resonators’ drug delivery efficiency is at least twice that of only one resonator.
