Multicomponent reactions(MCRs)can construct complex molecules from inexpensive and readily available small organic molecules,which are characterized by simple operation and high selectivity.This method conforms to an ideal reaction mode of green chemistry,and has become a hotspot for scientific researchers by its advantages of broad applicability.Compounds containing pyrrole structures are abundant in natural products and have a wide range of biological activities.They are widely used in the fields of drug synthesis and functional materials.In view of this,seeking new ideas or new routes for the synthesis of pyrrole derivatives has attracted the attention of pharmacologists and synthetic chemists.Tetramic acid(2,4-pyrrolidinedione)is a class of pyrrole-containing compounds with biological activities,also known as 1,3-dicarbonyl compounds.Based on green chemistry,an efficient and clean method was developed to synthesize a series of pyrrole derivatives under the design of “one-pot” multi-component reaction synthesis strategy and 37 unreported pyrrolo heterocycles were synthesized.Finally,the biological activities of pyrrole derivatives were explored and expected to provide a reference for specific drugs.Four types of of 5-substituted pyrrolidine-2,4-diones were prepared by using the monoethyl malonate acid chloride and different amino acid ethyl ester hydrochloride as raw materials through Dickman ester condensation reaction and decarboxylation reaction.29 new pyrrolopyrans compounds were synthesized using chiral tetramic acid with benzaldehyde derivatives and malononitrile as substrates via “one-pot” three-component reaction.The reaction is highly efficient with DMAP as catalyst in mild absolute ethanol.8 unreported pyrrolopyridine derivatives were synthesized using chiral tetramic acid,substituted benzaldehyde,ammonium acetate and ethyl acetoacetate as substrates via “onepot” four-component reaction.The reaction performed easily in ethanol with cheap sodium bicarbonate as catalyst.19 kinds of pyrrole derivatives were tested on the antitumor activity of human colon cancer cells,and showed good effects,which provided a new idea for the research of drugs.Figure 59;Table 17;Reference 115...
