The skin is an important barrier that protects the body from the outside world and a healthy and intact skin is important for human safety and health.However,as an organ in direct contact with the external environment,the skin is prone to injury and wounding in emergency and accidental situations,and bleeding and wound infection caused by wounds can be a serious risk to human health and safety,so it is important to develop wound dressings for the treatment of bleeding wounds.Currently many researches on wound dressings often suffer from a single function that cannot cope with a wide range of wound conditions,complex preparation processes and high costs.Therefore,it is important to develop a wound dressing that is hemostatic,antibacterial and can accelerate wound healing at the same time,and which is simple and inexpensive to prepare.In this study,a wound dressing combining hemostatic,antibacterial and wound healing functions was prepared by freeze-drying using chitosan,graphene oxide and egg white as raw materials.Quaternized chitosan was modified by quaternization,and quaternized chitosan/graphene oxide(QCS/GO)composite wound dressing was prepared based on the electrostatic interaction between quaternized chitosan and graphene oxide using freezedrying method.The hemolysis rate of the QCS/GO dressing is only 3.4%,which is lower than the safety standard of 5% and can be regarded as non-hemolytic.SEM of cell adhesion showed the normal physiological morphology of the immobilized cells,demonstrating the good blood and cytocompatibility of the composite dressing.The results of in vivo and in vitro hemostasis tests validate the excellent hemostatic properties of the composite dressing.Quaternized chitosan/graphene oxide/egg white protein(QCS/GO/EW)composite wound dressing was prepared by introducing egg white protein based on quaternized chitosan and graphene oxide.The antimicrobial results showed that the QCS/GO/EW composite wound dressing inhibited the growth of Staphylococcus aureus and Escherichia coli,and the dressing had excellent antimicrobial effect;the hemostasis experiments in mouse tail break model and mouse liver injury model demonstrated that the QCS/GO/EW dressing could stop bleeding faster and reduce blood loss compared with gauze and haemostatic cotton,and had excellent hemostatic performance.The CCK-8 test showed that QCS/GO/EW dressing was not cytotoxic and the fluorescence micrographs showed that QCS/GO/EW dressing promoted the proliferation of L929cells;further wound healing experiments in a mouse model of total skin defect demonstrated that QCS/GO/EW dressing accelerated wound healing.
