Both gynostemma pentaphyllum and honey are precious natural products for medicinal and edible purposes,which have become research hotspots in the development of functional food in recent years.In this study,gynostemma pentaphyllum and honey were used as research materials to explore the extraction process conditions of gynostemma saponin and optimized the dection method for saponin of gynostemma pentaphyllum.Subsequently,paste of Gynostemma and honey was prepared to explore its mechanism of action on hyperlipidemia mice.This study was divided into four chapters and major content are as follows:(1)Gynostemma saponins were extracted by hot reflux with 75%ethanol,and the extract was purified by D-101 macroporous adsorption resin.The optimal extraction process was determined by single factor experiments and response surface experiment.And the recovery rate of total saponins of Gynostemma pentaphyllum reached 42.197 mg/100 g with the ratio of solid to liquid 1:50(g/m L),the treatment temperature of 80°C,and the extraction time of1 h.The influence of factors ranked as:solid-liquid ratio>extraction time>extraction temperature.(2)The contents of total saponin and monomer saponins were respectively determined by the optimized vanillin-perchloric acid colorimetric(VPAC)and high-performance liquid chromatography(HPLC),and the Gynostemma honey paste was prepared by mixing the water extract of Gynostemma saponins with honey in a certain proportion.The results showed that ginsenoside Rb1 had a maximum absorption at 550 nm,with a good linear range of 0.04-0.2 mg,and the correlation coefficient was r~2=0.9997,the average sample recovery rate was 102.65%,the relative standard deviation(RSD)was 2.77%(n=6).and the extraction and purification of Gynostemma saponins using D-101 macroporous adsorption resin was simple,accurate,high sensitivity and good repeatability,which can be used for the determination of total saponins in Gynostemma pentaphyllum.(3)To study the effect of gynostemma honey paste on high-fat and high-sugar diet-induced hyperlipidemia in mice.The monosaponin content of gynostemma extract and its honey paste were analyzed by HPLC-DCD,and the genes associated with the formation of hyperlipidemia in mice were determined by Q-PCR to explore the mechanism related to the alleviation of hyperlipidemia by gynostemma honey paste.The results showed that gynostemma honey paste contained monosaponin XLIX and A at 0.055 mg/g and 0.01 mg/g,respectively.10 g/kg of gynostemma honey paste significantly reduced body weight,fasting blood glucose,liver index,serum and liver total cholesterol(TC)levels,increased glutathione peroxidase(GSH-Px)activity in liver tissue,and decreased gynostemma honey paste intervention also attenuated the expression of chronic metabolic inflammatory genes(IL-6,NF-κB)induced by high-fat diet.This study showed that gynostemma honey paste had a good hypolipidemic effect on mice,which provides a basis for the development of gynostemma functional food.
