time,it can cause gout.So far,drugs that can completely cure hyperuricemia and gout have not been developed.The therapeutic drugs used clinically(such as allopurinol and febuxostat)are very effective xanthine oxidase inhibitors,but these drugs There have been reports of many side effects,damage to organs,various diseases,and even death.Therefore,today when the safety of existing therapeutic drugs is questioned,there is an urgent need for alternative strategies for the treatment of hyperuricemia without any side effects.Therefore,many scholars are studying xanthine oxidase inhibitors derived from natural products in order to achieve high safety.,Natural product active ingredients with low toxicity or no toxic side effects have become an effective method for the treatment of hyperuricemia.In this paper,on the basis of the polysaccharide extract of Inonotus obliquus in the early stage of the laboratory,the inhibitory effect of the crude polysaccharide of Inonotus obliquus and its three graded alcohol precipitation components(IOP40、IOP60 and IOP80)on XO in vitro was studied.By comparing the half inhibitory concentration(IC50)of the alcohol precipitation component on XO,and further explore the inhibitory mechanism and type of inhibition of the component with the strongest inhibitory activity on XO;the mouse hyperuricemia model was induced by instilling potassium oxazinate for 7 days to explore The effect of Inonotus obliquus polysaccharide on the uric acid level,organ function damage and oxidative stress response in hyperuricemia mice.The histological morphology of mouse kidneys was observed to explore whether Inonotus obliquus polysaccharide is in the animal body.It has kidney protection effect;the basic formula of Inonotus obliquus polysaccharide composite solid beverage was studied through single factor test and orthogonal test.According to the sensory evaluation,determine the effect of white sugar,non-dairy creamer,almond powder,coffee powder,green tea powder and Inonotus obliquus polysaccharide on the taste and further optimize to determine the best addition amount.The best effect is selected through single factor experiment.In order to prevent the solid powder from absorbing water and agglomerating,a single factor experiment is used to determine the optimal addition amount of the anti-caking agent.The main research conclusions are as follows:(1)Through the in vitro enzyme inhibition experiment of Inonotus obliquus crude polysaccharide and its graded alcohol precipitation polysaccharide on XO,the four samples have a certain inhibitory effect on the activity of XO,and the IC50value is calculated to screen out the effect of XO.The most inhibitory polysaccharide components are IOP40 and IOP60,and their IC50values are 0.324±0.02 mg/m L and 0.386±0.01mg/m L,and there is no significant difference.The reversible effects of IOP40 and IOP60on XO inhibition and the types of inhibition were investigated respectively,and the results showed that they are both effective reversible XO inhibitors,and the inhibition types are all mixed inhibition types.(2)Through intragastric administration of different doses of Inonotus obliquus alcohol precipitation polysaccharide component IOP60,the experimental results show that:IOP60 has the ability to reduce the level of uric acid in the body,and it can also reduce the serum level of mice to a certain extent.The content of BUN and Cr.After further observation of the mouse kidney tissue morphology,it was found that the renal disease was alleviated under the action of IOP60,indicating that IOP60 can play a certain protective effect on the kidney and can significantly alleviate the damage caused by high levels of uric acid to the kidney.In addition,IOP60 also has the ability to inhibit XO,a key enzyme in the pathway of uric acid metabolism,which can fundamentally reduce high levels of uric acid.According to the determination of SOD and CAT activity and MDA and GSH content in each group of mice,it was found that high-dose IOP60 can effectively increase the activity of SOD and CAT,and has a good regulatory effect on the levels of GSH and MDA,proving that IOP60 has Strong anti-oxidant ability,thereby reducing the occurrence of oxidative stress damage in the body,and has an anti-inflammatory effect.(3)Through the research on the flavor of the Inonotus obliquus composite solid beverage,the Inonotus obliquus extract,white sugar,non-dairy creamer and other ingredients are compounded into a new type of plant solid beverage,and the optimal formula is determined to be 50%of white sugar,26%of non-dairy creamer,1.5%of coffee powder,1.5%of green tea powder,9%of Inonotus obliquus extract,10%of almond powder.According to this formula,the flavored drink has the best taste.Through single factor test,it was determined thatβ-cyclodextrin was used as the stabilizer of the solid beverage,the addition amount was 0.20%.The optimal addition amount of anticaking agent silicon dioxide was 0.12g/10g.On this basis,the product standard of Inonotus obliquus composite solid beverage was established. |
