Hyperuricemia(HUA)is a metabolic disorder caused by purine metabolism disorder.Xanthine oxidase(XOD)is an important target for the prevention and treatment of HUA,and it can participate in the production of uric acid.Persistently high levels of uric acid can cause HUA,which in turn can lead to gout.Regulating human health through diet is more and more recognized by consumers,so some natural active products with uric acid lowering or anti-gout activity are favored.The tender stems of Artemisia selengensis Turcz(AST)is usually eaten as vegetables,but a large number of leaves of AST belong to waste,improper disposal will pollute the environment,and studies have shown that the leaves have antioxidant and XOD inhibition activities,and have the potential effect of lowering uric acid.Therefore,the comprehensive development of AST is not only beneficial to increase the by-value of artemisia scoparia products,but also meets the development needs of resource-conserving society.For this purpose,three aspects are studied:first,the nutritional composition,antioxidant capacity and XOD inhibitory activity of different AST are studied;Second,AST leaves were used as the main raw materials,supplemented by Gardenia jasminoides Ellis,celery seed,Rubus chingii Hu,Siraitia grosvenorii and other raw materials to make AST compound drinks;Third,to evaluate the effects of AST compound drinks on uric acid and liver and kidney functions in acute HUA mice induced by potassium oxazinate,in order to provide a basis for the development of new functional AST products.The main results are as follows:(1)Nutrient composition and functional activity of AST of different varieties were compared.Principal component analysis was used to construct a mathematical evaluation model for potential uric acid lowering effect of AST.The 8 indexes were simplified into one principal component,and its cumulative variance contribution rate was 93.384%,which could reflect most of the quality information of AST.The results showed that the antioxidant capacity,nutrient content and XOD inhibition rate of No.8 were the highest,and the comprehensive score of principal component analysis was 4.7820,which could be used as raw material for subsequent experiments.(2)Orthogonal experiment and response surface analysis were used to optimize the extraction and compounding process of AST.The results showed that the optimal extraction conditions were as follows:solid-liquid ratio of 1:40(m/v),time of 40min,temperature of 95℃.Under these conditions,the XOD inhibitory rate of the extract was the highest(74.21%).The optimal supplemental levels of the excipients were Gardenia jasminoides Ellis extract 0.6%,celery seed extract 1.0%,Rubus chingii Hu extract 1.4%,Siraitia grosvenorii extract 0.6%,and the XOD inhibition rate was 85.30%.With xylitol 13%,citric acid 0.09%,β-cyclodextrin 0.6%,sensory score was optimized to obtain the best formula of AST compound drink.(3)The HUA mouse model was induced by oteracil potassium,AST compound drink was used as the test object,allopurinol was used as positive control,and related indexes in serum,XOD activity in liver and liver and kidney index of mice were determined,and the intervention effects on uric acid and liver and kidney functions of HUA mice were studied.The results showed that the AST compound drink can reduce model mice serum UA,Cr,BUN,AST,ALT levels and XOD content in the liver,and combined with liver and kidney biopsy results,show that the compound drink can play a role of good drop uric acid,and does not affect the animal’s liver and kidney function.
