| Human skin wounds are easily invaded by bacteria and microorganisms during the healing process and cause inflammation.Inflammation can slow recovery and cause repeated suffering.Since the nanofiber morphology is similar to that of natural extracellular matrix(ECM),nanofibrous membranes with antibacterial properties can be prepared by electrospinning method and used in wound healing process.Generally,the preparation of nanofiber membranes is separated from the application process,which reduces the use effect of many volatile and perishable drugs,and the adhesion of the fibrous membranes is unsatisfactory,which is likely to cause secondary damage to the wound during use.In situ electrospinning can deposit electrospun fibers directly to the wound,which can effectively improve the problem of electrospun fiber dressings.Four kinds of nanofibrous membranes with antibacterial properties were prepared in situ by a portable hand-held electrospinning device: PVB/Berberine(BB),PCL/PVA/PCL-Bupleurum(BL),Zein/clove essential oil(CEO),Zein/cinnamon essential oil(CO).The prepared nanofiber membranes were tested by SEM,FTIR,pore size,hydrophilicity and hydrophobicity,air permeability and antibacterial.Discover:(1)PVB/BB membrane is hydrophobic.After adding BB,the fiber diameter is reduced.The antibacterial property of nanofiber membrane with BB content of 5 wt% is the best,the pore size is 4.0502 ± 0.13 μm,and the air permeability is 164 ± 64.2 mm/s.(2)The PCL/PVA-BL/PCL membrane exhibited slow hydrophilic properties and could fully absorb wound exudate.When the BL content was 5 wt%,the nanofiber membrane had the best antibacterial properties,with a pore size of 2.143 ± 0.33 μm and an air permeability of 61.8 ± 16.7 mm/s.(3)Zein/CEO membrane is hydrophilic and can fully absorb wound exudate.After adding CEO,the average fiber diameter decreased The nanofiber membrane has good antibacterial properties,with a pore size of 8.8 ± 0.67 μm and an air permeability of168.2 ± 43.3 mm/s.After 24 hours,the cell viability was higher than 90%,and animal experiments showed that the wound was almost completely closed after 11 days of in situ electrospinning of the fibers to the wound.(4)Zein/CO membrane is hydrophilic and can absorb wound exudate.After CO addition,the fibers were finer and more uniform.The nanofiber membrane has good antibacterial properties,the pore size is 7.5 ± 0.72 μm,and the air permeability is 76.1 ±5.45 mm/s.Animal experiments demonstrated that the fibrous membrane was prepared and deposited in situ at the wound,and the wound was nearly closed after 11 days.Experiments have shown that the electrospun fiber membranes added with plant-derived antibacterial substances have different degrees of inhibition on Gram-positive and negative bacteria,and in-situ electrospinning can directly deposit them into wounds to form dressings,reducing inflammation.Thereby effectively promoting wound healing. |
