Peach gum,a colloidal exudate of Rosaceae plants such as peach,is traditionally used as medicine and health food.Upon mechanical damage or microbial infection,peach gum will be exuded to limit the spread of pathogens and protect the tree.Peach gum has a wide range of applications in food,cosmetics and other fields.The main component of peach gum is macromolecular polysaccharide,which is difficult to dissolve in water.Here we first prepared peach gum polysaccharide with good solubility using high pressure method,and then,according to its protective properties in nature,studied its potential wound healing effect,physicochemical and rheological properties,and application in skin creams.The main results are as follows:1.Potential wound healing effect of peach gum polysaccharide.Peach gum has a protective and repairing effect on the trunk.Therefore,we first selected mouse fibroblast L929 to explore the potential wound healing effect of peach gum polysaccharide by cell scratch assay,and found that the polysaccharide could promote the migration of L929 cells to the scratch area,with a gap closure rate of 56%,which was 1.5 times that of the control,indicating a potential wound healing effect of peach gum polysaccharide.2.Antioxidant and anti-inflammatory activities of peach gum polysaccharide.Oxidative stress and inflammatory responses are closely related to wound healing.Therefore,we further investigated the antioxidant and anti-inflammatory effects of peach gum polysaccharide.First,we analyzed the effects of peach gum polysaccharide on H2O2-induced L929 cells by flow cytometry,and found that the polysaccharide could reduce the level of reactive oxygen species(ROS),upregulate mitochondrial membrane potential and improve G1 phase cell cycle arrest,demonstrating the antioxidant activity of the polysaccharide.Then,we determined the effect of peach gum polysaccharide on nitric oxide in lipopolysaccharide-induced inflammation model using RAW264.7 cells,and found that peach gum polysaccharide had the effect of reducing nitric oxide content,indicating the anti-inflammatory properties of peach gum polysaccharide.3.The physicochemical and rheological properties of peach gum polysaccharide.Based on the above work,we then analyzed the molecular weight distribution,monosaccharide composition,infrared spectrum and rheological properties of the bioactive peach gum polysaccharide.High performance gel permeation chromatography revealed a single peak in the molecular weight distribution of the polysaccharide,with a molecular weight of 60.3 kDa.The monosaccharide composition of the peach gum polysaccharide was arabinose,galactose,xylose,glucuronic acid,mannose and rhamnose,with molar percentages of 57.4%,25.9%,10.9%,2.2%,1.9%and 1.7%,respectively,as determined by gas chromatography analysis.Infrared spectrum analysis showed typical characteristic absorption peaks of the peach gum polysaccharide.Rheological analysis indicated the shear thinning phenomenon of the peach gum polysaccharide,i.e.decreased viscosity with the increase shear rate,indicating that the peach gum polysaccharide solution was a pseudoplastic non-Newtonian fluid.4.Application of peach gum polysaccharide in cosmetics.The above findings suggest that the peach gum polysaccharide has potential wound healing effect as well as antioxidant and anti-inflammatory activities,and the shear thinning phenomenon indicates good smearing and absorption properties.We measured the effect of peach gum polysaccharide on rheological properties and skin feeling in experimental creams with.Rheological analysis showed that the peach gum polysaccharide was able to improve the viscosity,flow properties and gel structure of the cream.The creams with or without the polysaccharide showed similar transparency,moisturizing feeling,smoothness and radiance,while the cream with peach gum polysaccharide had better freshness and firmness.In summary,peach gum polysaccharide has potential wound healing effect and antioxidant and anti-inflammatory activities,and its aqueous solution is a pseudoplastic non-Newtonian fluid.When applied to cosmetics,the polysaccharide can improve the rheological properties and skin feeling of the cream. |
