The peach gum is the gum substance secreted from the bark of rosaceae such as Prunus persica?L.?Batsch or Prunus davidiana?Carr.?Franch,which has the effect of relieving pain,replenishing blood and qi,and treating the symptom of fever and thirsty,just like diabetes.As well,it occupies the advantages of non-toxicity,large yield and low price.Now the peach gum has been widely used in food and health care.But its basic research is still relatively weak,lacking of quality control standards.And the peach gum yield is big in our country,so application value still needs to dig further.In this paper,from these aspects,the main substance polysaccharide of the peach gum was reserached,including structure characteristics and activities to verify its efficacy in promoting immunity and lowering blood glucose in traditional Chinese medicine,and provide experimental basis for clinical application and product development.Based on the active components of peach gum,the quality control was researched.The peach gum polysaccharide has the anti-oxidation and moisturizing effects,on this basis we developed the peach gum hand cream for further excavating the peach gum application value,and enhanced its utilization rate.The full text includes the following five parts:Section 1 Overview the application in food and medicine of peach gumThe source and food,medicinal value of the peach gum was summarized.Changes of metabolism in plants during the formation of peach gum was summarized.Classification and chemical,physical characteristics of peach gum was summarized.Pharmacological activity value of peach gum was introuduced.As well as the application in the medical,food,skin care,preservative materials and other aspects was introuduced.Section 2 Separation and composition analysis of peach gum polysaccharides?PGP?Peach gum polysaccharides?PGP?were obtained by hot water extraction and ethanol fractional precipitation,and then purified by elution,DEAE-52 cellulose column,dialysis desalination and Gel-100 molecular sieve column chromatography.The relative molecular mass of PGP was 1.11×103 KDa detected by HPGPC.HPLC-ELSD method was used to detect its monosaccharide composition.The results showed that PGP was composed of xylose,arabinose,glucose and galactose,with a molar ratio of 5.26:38.86:5.95:49.92.Infrared spectrum analysis and methylation experiment proved that there is pyran ring in this structure,and the connection mode were?1?6?-galactose,1-arabinose.Section 3 The activity of increasing macrophage proliferation and inhibiting a-glucosidase of PGPPlant polysaccharic s often have good effects on control the blood glucose and enhancing immunity.In this study,M'.T assay and a-glucosidase inhibition assay were performed to verify the activity of PGP.The resu showed that PGP can promote proliferation of macrophages in the abdominal cavity of mice.When the concentration of PGP was 125 ?g/mL,the proliferation rate was 137.16%,compared with the blank group.At the same time,when the concentration reached 4.13 mg/mL,the inhibition a-glucosidase enzyme rate was 50%,but the maximum inhibition rate was only 70%.Section 4 Quality control method of peach gumThe fingerprint of peach gum was established.The similarity of peach gum in 10 different areas was more than 0.96,and it was obviously different from fake gelatin.The common pattern of peach gum's infrared fingerprint was found.A fast and lossless identification method was established.The type and content of monosaccharide in peach gum were determined by hydrolysis of 2M trifluoroacetic acid.The hydrolysis conditions of peach gum by 2M trifluoroacetic acid were optimized through single factor experiment,and the conditions were 105? and 2 h.The HPLC-ELSD method was established.Under this condition,the determination of peach gum in 10 different areas showed that the monosaccharides in these 10 different areas were the same,but the content is different.The highest content was galactose,whichwas 305.33?358.99 mg/g,followed by arabinose 262.04?315.13 mg/g.The total sugar content in shengzhou,cangnan,zhejing and wuxi,Jiangsu were significantly higher than that in suizhou,hubei and bijie,guizhou.Based on the relationship of the peach gum formation process and rainfall,we speculated that the results may be related to the climatic rainfall.The method was repeatable,accurate,linear,precise and stable can be applied to the quality control of peach gum.The peach gum was qualitatively identified and distinguished from other counterfeits by infrared fingerprint,and the types and contents of monosaccharides in peach gum were determined after acid hydrolysis.The authenticity and quality control of peach gum were analyzed qualitatively and quantitatively.Section 5 Study on preparation of peach gum hand creamThe peach gum was used in skin care products,which is a rare environment friendly,moisturizing,antibacterial,antioxidant and natuarl materials.In order to further explore the application value of peach gum,we developed the peach gum hand cream.Firstly,the moisture absorption,moisture retention and oxidation resistance of peach gum were evaluated.The results showed that the peach gum had better moisture retention effect than sodium alginate,glycerin and hyaluronic acid.The scavenging efficiency of ABTS+ was 50%,when the concentration of crude polysaccharides was 6.7mg/mL.And the scavenging efficiency of ·OH was 50%,when the concentration of crude polysaccharides was 13.7 mg/mL.The optimum emulsifying temperature and time of hand cream were optimized by single factor experiment,and condition was 80? and 20 min.L27(313)orthogonal experiment was designed to selected the optimal formula.The optimal formula was:cetacean stearin 0.5%,lecithin 0.0675%,horoba oil 0.4%,pewter 1%,dimethyl silicone oil 0.225%,glycerin 3%,squalane 0.225%,carbomer 0.04%,coconut glucoside 0.4%,taojiao 0.3%,monoglyceride 0.09%,hyaluronic acid 0.0275%,stearic acid 0.15%.The products with the optimal ratio were tested according to the industry standards for sensory and physicochemical indexes.And they all up to standars.
