Metabolic syndrome is a high incidence disease in adults,which seriously endangers human health.Drugs currently used for the treatment of metabolic syndrome generally have large side effects and high prices.The medicinal and edible homologous plants have good safety and diverse biological activities,and have good potential for treating metabolic syndrome.This paper explores the effect and mechanism of Pu-erh tea compounded with mulberry and dendrobium in the intervention of metabolic syndrome.The research work mainly includes the following aspects:First,the effective components of Pu-erh tea,mulberry leaves,mulberry branches and dendrobium were separated and extracted and their contents were analyzed.The content of theabrownin in the pigment of Pu-erh tea was 6.47%,which was higher than 0.27% of theaflavin and 1.91% of thearubigin.,is the main tea pigment in Pu-erh tea extract.Mulberry leaves and mulberry branches mainly contain flavonoids,and the total flavonoids are 15.70%and 34.00%,respectively.The main components are hyperin,rutin and astragaloside by LC-MS/MS comparison with standard products.The total polysaccharide of Dendrobium is the main component of Dendrobium,which is 45.00 ± 5.00% in the extract of Dendrobium.Secondly,the composition of Pu-erh tea compounded with mulberry leaves,mulberry branches and dendrobium extracts was screened.Through in vitro experiments,it was found that compound Pu-erh tea increased the inhibitory effect on α-glucosidase activity compared with the single-use group of Pu-erh tea.And through the lipid accumulation model of oleic acid-induced AML-12 cells,it was found that compound Pu’er tea could significantly reduce lipid accumulation in cells compared with the single group.Two compositions of compound Pu’er tea were determined(Pu’er tea: mulberry leaf/mulberry branch: dendrobium = 5:2:2),named as Pu’er tea complex 1 and Pu’er tea complex 2.Thirdly,the pharmacodynamic effects of two compound Pu-erh teas on metabolic syndrome mice were explored.The results show that Pu’er tea complex 1 can reduce the body’s blood sugar level,enhance insulin sensitivity,and improve hyperglycemia in mice with metabolic syndrome;Pu’er tea complex 2 can prevent adipocyte hypertrophy and reduce total cholesterol content,and improve hyperlipidemia in mice with metabolic syndrome;Pu’er tea complex 1 and 2 can prevent hepatocyte enlargement and hepatocyte necrosis,reduce fat vacuoles,and improve non-alcoholic fatty liver disease in mice with metabolic syndrome.The effects of the two compound Pu’er teas in improving metabolic syndrome in mice were significantly better than those of the single-use group of Pu-erh tea.Finally,the mechanism of Pu’er tea complex 1 intervening in metabolic syndrome was studied.Through the analysis of microbial diversity,it was found that compound Pu’er tea 1could increase the abundance of beneficial bacteria in the intestine,inhibit the abundance of harmful bacteria,and increase the abundance of short-chain fatty acid,suggesting that Pu’er tea complex 1 can improve metabolic syndrome through the mechanism of intestinal flora;the effects of Pu’er tea complex 1 on key proteins and signaling pathways in mouse liver were studied by real-time quantitative PCR and Western Blotting techniques,and it was found that Pu’er tea complex 1 can significantly increase the phosphorylation of AMPK,down-regulate the key protein of SREBP1,promote fatty acid oxidation(CPT1A),and inhibit fatty acid synthesis(FAS),which are the main mechanisms for improving lipid metabolism disorders in mice with metabolic syndrome.In this paper,it is found that the combination of Pu’er Tea,Mulberry Leaf and Dendrobiumthe can regulate the glucose and lipid metabolism disorder caused by metabolic syndrome through various effects,which provides a theoretical research basis for the development of safe and effective drugs for improving metabolic syndrome with few side effects. |
