| Andrias davidiauns has large-scale breeding in Wuyuan,Jiangxi Province,and it is in urgent need to study its deep processing to produce high-value food.In this paper,the muscle was processed into Andrias davidianus meat peptide(ADMP)by enzymatic lysis to study its in vitro antioxidant activity and stability,establish cell oxidative dam-age model,further explore the possible mechanism of action,and ADMP was devel-oped into a functional food with antioxidant function.In order to clarify the effect of enzymatic conditions on ADMP,the clearance of1,1-diphenyl-2-trinitrohydrazine(1,1-Diphenyl-2-picrylhydrazyl radical,DPPH)under different conditions were studied.The results showed that the trypsin and papain were combined,the process conditions were 5 h,the enzyme amount was 0.25%,the mate-rial ratio was 1:6(g:m L),and the enzyme temperature was 50℃,and the DPPH clear-ance was 90.16%.This analysis showed uniform amino acid distribution,high nutri-tional value and molecular weight concentrated below 2 k Da.After isolation and puri-fication of ADMP,the DPPH with small molecular weight cleared through the gel chro-matcolumn,four components were separated,and the components with the strongest DPPH clearance reached 98.24%.Optimize the enzymatic hydrolysis process to pro-vide a deep layer of basic research and product development preparation data.ADMP has antioxidant function and studied its antioxidant activity and stability in vitro.The results showed that ADMP has good antioxidant activity,reducing force,and can scavenge DPPH radicals,superoxide anion radicals and hydrox(Oxhydryl,·OH)radicals.ADMP has good heat resistance;can maintain good antioxidant activity in neu-tral environment,adding sucrose,glucose and xylose reduces antioxidant activity,Na Cl;preservative sodium benzoate affects antioxidant stability;metal ions,Cu2+and Zn2+;ADMP decreases DPPH radical clearance after pepsin,with the same trend after trypsin digestion.In industrial processing and transportation,the quality of ADMP is improved by improving the environmental conditions and controlling the impact of adverse fac-tors on ADMP to meet the needs of actual production.In vitro oxidative damage cell model was established to study the antioxidant prop-erties and mechanism of ADMP.arrival at a conclusion,In the Hep G2 cells,ADMP can reduce hydrogen peroxide(Hydrogen peroxide,H2O2)caused by the cytotoxicity,Can reduce the intracellular reactive oxygen species(Reactive oxygen species,ROS)horizontal,Improved superoxide dismutase(Superoxide dismutase,SOD)activity,cat-alase(Catalase,CAT),glutathione peroxidase(Glutathione peroxidase,GSH-PX)live enzyme,glutathione(Glutathione,GSH),malondialdehyde(Malondialdehyde,MDA)content,Effective for removing peroxygenic free radicals within the cell,To attenuate the intracellular levels of oxidation,It acts to protect the cells.According to the results of protein blotting(Western blot,WB),ADMP downregulated the expression of apop-tosis-related proteins such as Caspase-3,Bcl-2-Associated X,Bax),anti-apoptotic pro-tein B-cell lymphoma-2(Bcl-2),inhibited the apoptosis of Hep G2 cells,and played a protective role against oxidative damage.According to the results of antioxidant exper-iment,a functional product of ADMP was developed.The formula is ADMP,sea cu-cumber peptide,oyster peptide,iso Vc sodium,inulin,oligofructose,mint essence,lemon essence,pure water.This article optimizes the formulation of oral liquid. |
