In recent years,the field of fluorescent probe has developed very rapidly.Compared with the traditional detection methods,fluorescent probe has the advantages of good selectivity,high selectivity and easy operation,so it has become a better choice for the detection of analytes.Especially in the detection of metal ions and other anions,and the probe with low biological toxicity can also be used in cells,animals and plants,which has great application value.Mercury ion pollution does great harm to human health.Especially when inorganic mercury is transformed into organic mercury,it is more toxic,which will cause brain injury,body organ injury,behavior disorder and even death.Copper ion is an important component of trace elements in human body,but when a large amount of copper is accumulated in human body,it is very easy to harm the body and cause a series of health problems,such as liver cirrhosis,cancer and other diseases.In recent years,studies have shown that excessive accumulation of copper ions can lead to Parkinson’s and Alzheimer’s disease.Based on this,we designed and synthesized four fluorescent probes to detect mercury ion and copper ion respectively.We not only studied the optical properties of the probes,but also further explored the practical application of the probes.The main contents of this study are as follows:Based on the recognition mechanism of fluorescent probes and the relevant research and exploration of scientific researchers,four probes of CNS,HQ1,HBD and DLX were designed and synthesized with coumarin derivatives as fluorop Hores.The structures of the synthesized probes were characterized and analyzed by 1H NMR,13C NMR and HRMS to determine that the structure of the final synthesized compound is consistent with that of the target product we designedThe probe CNS can detect mercury ions by deprotection reaction with mercury ions.The probe has good selectivity,high sensitivity and good anti-interference effect.It can be used to detect mercury ions in zebrafish and its biological toxicity in vivo.Because the probe shows the effect of fluorescence enhancement when detecting mercury ions,it can be detected not only qualitatively but also quantitatively.Probes HQ1,HBD and DLX are fluorescent probes designed and synthesized for copper ions.These three probes have acylhydrazone structure.Using the excellent coordination ability of acylhydrazone structure,they complex with copper ions to form stable complexes,so as to achieve the purpose of detecting copper ions.The probe has specific detection for copper ions,is not disturbed by other metal ions,has fast response time and good stability.It has low biological toxicity and can be used to detect copper ions in cells and zebrafish.
